fa بررسی همراهی r‌s1421085 و rs9939609 در ژن FTO با دیابت دوران بارداری در شهر اراک Association Study of rs1421085 and rs9939609 in FTO Gene with Gestational Diabetes in the Arak City علوم پایه Basic Sciences پژوهشي اصیل Original Atricle مقدمه: دیابت بارداری به عنوان هایپرگلیسمی با شروع یا اولین تشخیص در دوران بارداری، با فراوانی کمتر از دیابت عمومی تعریف می‌شود. اضافه وزن و چاقی، سن بالا در زمان باروری، سابقه قبلی دیابت بارداری، سابقه خانوادگی دیابت نوع 2 و نژاد، از عوامل خطر دیابت بارداری است. اخیراً ارتباط بین دو پلی‌مورفیسم ژن FTO (rs9939609، و rs1421085) و خطر دیابت بارداری در مطالعات با نتایج متناقض بررسی شده است. هدف ما از مطالعه حاضر، بررسی همراهی ژنتیکی دوrs1421085  و rs9939609 از ژن FTO بر روی دیابت بارداری بود. روش کار: در این مطالعه موردی- شاهدی، خون محیطی به میزان 5 سی‌سی از 100 فرد مبتلا به دیابت بارداری و 116 خانم باردار شاهد از بیمارستان طالقانی شهر اراک اخذ و سپس DNA استخراج شد. پس از طراحی پرایمر‌های اختصاصی، با روش Tetra ARMS-PCR ژنوتیپ‌ها را مشخص شد. اطلاعات دموگرافیک و ژنوتیپ‌ها بین دو گروه با استفاده از نرم‌افزار SNPSTAT مقایسه شد. یافته‌ها: همراهی معنی‌داری 0001/0 = P بین شاخص‌های چاقی، (Body mass index) BMI قبل و حین بارداری، سطوح HbA1c، FBS، کلسترول، تری‌گلیسرید در بین مادران مبتلا به دیابت باداری نسبت به مادران باردار سالم مشاهده شد. با توجه به تجزیه و تحلیل داده‌ها، همراهی معنی‌داری بین دو  rs1421085 و rs9939609 از ژن FTO با دیابت دوران بارداری مشاهده نشد. نتیجه گیری: با توجه به نتایج متناقص بین همراهی پلی‌مورفیسم‌های ژن FTO با دیابت بارداری جهت شناسایی و درک بهتر پاتوژنز بیماری مطالعات بیشتری با تعداد نمونه بالاتر و گروه‌های نژادی بیشتر نیاز است. با شناسایی پروفایل و جایگاه‌های استعداد دهنده ژنتیکی به دیابت بارداری در آینده می‌توان جهت مدیریت بیماران استفاده کرد. Introduction: Gestational diabetes is defined as hyperglycemia with onset or first diagnosis during pregnancy, with a lower frequency than general diabetes. Overweight and obesity, old age at the time of pregnancy, previous history of gestational diabetes, family history of type 2 diabetes, and race are risk factors for gestational diabetes. Recently, the relationship between two polymorphisms (rs9939609 and rs1421085) of Fat mass and obesity-associated (FTO) gene and the risk of gestational diabetes has been investigated in some studies with conflicting results. Therefore, our aim of the present study was to investigate the association study of rs1421085 and rs9939609 of the FTO gene on gestational diabetes. Methods: In this case-control study, five cc of peripheral blood was collected from 100 cases with gestational diabetes and 116 control pregnant women from Taleghani Hospital in Arak, and then DNA was extracted. After designing the specific primers, the genotypes were determined by the Tetra ARMS-PCR method. Demographic information was compared with genotypes between the two groups were compared using SNPSTAT software. Results: A significant correlation was observed between obesity, BMI before and during pregnancy, HbA1c, FBS, cholesterol, and triglyceride levels among mothers with gestational diabetes compared to healthy pregnant mothers P = 0.0001. According to data analysis, there was no significant association between rs1421085 and rs9939609 of the FTO gene with gestational diabetes. Conclusions: Considering the conflicting results between the association of FTO gene polymorphisms with gestational diabetes, to better identify and understand the pathogenesis of the disease, more studies with a higher number of samples and more racial groups are needed.
