other توقف تکثیر در رده سلولی MCF_7 به دنبال استفاده از داوری سیس پلاتین Cell Proliferation Inhibition in MCF-7 Cell Line After Treatment With Cisplatin Drug روانپزشکی psychology پژوهشي اصیل Original Atricle <div style="text-align: justify;">مقدمه: یکی از شیوه هائی که در درمان سرطان مطرح است استفاده از داروهای شیمی درمانی می باشد. سرطان پستان، بیماری شایعی است که هنوز داروی شیمی درمانی نتوانسته آن را به طور کامل درمان کند. در این تحقیق، توقف تکثیر سلول هایMCF-7  که یک رده سلولی سرطان پستان انسانی است به دنبال استفاده از داروی ضد سرطان سیس پلاتین بررسی می شود. مواد و روش ها: در ابتداء پس از انجام شمارش سلولی با رنگ تریپان بلو تعداد یکسانی سلول به فلاسک ها ریخته شده و فلاسک ها به دو گروه شاهد و مورد تقسیم شدند. در گروه مورد یک میکرومول داروی سیس پلاتین به مدت یک ساعت و در گروه شاهد محلول سالین نرمال اثر داده شدند. سپس فلاسک ها به مدت 48 ساعت دیگر انکوبه شدند. پس از این مدت، در سلول های چسبیده دو گروه شاهد و آزمایشی به طور جداگانه با رنگ تریپان بلو شمارش سلولی انجام گرفت. همچنین سلول های چسبیده دو گروه شاهد و آزمایشی به طور جداگانه با تولوئیدن بلو رنگ آمیزی شده و با میکروسکوپ نوری مطالعه شدند. یافته ها: نتایج به دست آمده به دو گروه تقسیم شد. شمارش سلولی و میکروسکوپ نوری در شمارش سلولی نتایج به دست آمده به دو گروه تقسیم شد. شمارش سلولی و میکروسکوپ نوری در شمارش سلولی نتایج به دست آمده توسط ازمون آماری Mann-Withney  ارزیابی شد. نتایج نشان داد که تعداد سلول های چسبیده گروه ازمایشی نسبت به گروه شاهد کاهش معنی داری پیدا کرده است. در قسمت میکروسکوپ نوری، مشاهده شد که مورفولوژی سلول های چسبیده دو گروه شاهد و آزمایشی یکسان است. نتیجه گیری: از آنجا که سلول هایMCF-7  چسبیده در حقیقت سلول های در حال تکثیر هستند، بنابراین می توان گفت که داروی سیس پلاتین بدن تغییر در مورفولوژی سلول ها موجب توقف تکثیر سلول هایMCF-7  می گردد. </div> <div style="text-align: justify;">Introduction: One  of  the  methods  of  cancer  treatment  is  use  of  chemotherapic  drugs.  Breast  cancer  is  a  common  desease  that  chemotherapic  drugs  have  not  treated  it  yet. In  this  research, Cell  proliferation  inhibition  in  MCF-7  cells, a  human  breast  carcinoma  cell  line , after  treatment  with  Cisplatin, an  anticancer  drug  is  studied. Material  and  Method: At  first, after  cell  counting  with  trypan  blue  stain, equal  cells  were  added  to  flasks  and  flasks  were  divided  in  to  two  control  and  experimental  groups. In  experimental  group, 1  micro  mole  cisplatin  exposure  in  1  hour, where  control  group  with  normal  saline  solution. after  that, flasks  were  incubated  for  48  hours. After  passing  this  time, attached  cells  of  two  control  and  experimental  groups, were  stained  with  trypan  blue  for  cell  counting  and  with  toloiden  blue  for  light  microscopy  individually.  Obtained  results  were  divided  into  two  section, cell  counting  and  light  microscopy. In  cell  counting. Obtained  results  were  evaluated  with  Mann-Withney  Statistical  test. Results: results showed  that  number  of  attached  cells  in  experimental  group  was  reduced.  In  light  microscopy, morphological  features  of  attached  MCF-7 cells   in  two  groups  were  similar.
