Abstract

  Introduction: The increasing use of EMF (electromagnetic field) generating apparatus (refrigerators, computers, TV, etc) caused an increasing interest in investigations of its adverse effects on human health. This study is done to investigate the effects of EFM on Balb/c mice.

  Materials and Methods: This is an experimental study in which at first a circuit generating low frequency electromagnetic field (50 Hz, 15G) was designed. Then adult virgin female mice were placed in coil and exposed to 15 gauss electromagnetic field for 4 day and 6 hour per day. Then their blood was examined to assay the level of hormones (FSH, LH, estradiol, progesterone). Also ovary and uterus sections were studied with light & electronic microscope.

  Results : Results showed that the weight and size of ovary was not significantly affected in females exposed to the low frequency electromagnetic field and their offspring. Our results also showed that the number of ovary follicles were significantly affected in exposed females (p<0.05). Also the study of micrographs showed hetrochromatinated oocytes and follicular cells and increasing polysomes, accumulation of mitochondria and cleft nucleus. Decreasing amount of FSH, LH and 50% decrease in couplation rate was also seen as compared with the control group.

  Conclusion: Results of this study is indicator of EFM effects on gonads, structure and endocrine system and decreases fertility.

Abstract

Background: Hirsutism in women is identified as the increasing growth of terminal hair in areas of skin that are sensitive to androgen. The most common causes of hirsutism are PCOS (polycystic ovary syndrome) and idiopathic hirsutism. The role of insulin resistance and hyperinsulinemia as the most important pathogenesis mechanism of PCO is quite obvious, but there is not enough information on the role of insulin resistance in idiopathic hirsutism. Materials and Methods: This cross-sectional study was conducted on 16 to 40 year old women. Medical history and physical examination were completed for patients with PCOS and idiopathic hirsutism. Hormonal tests and sonography were run for both groups and insulin resistance was assessed using HOMA-IR formula. Data were analyzed using SPSS version. 17. Results: Mean of serum insulin levels in PCOS and idiopathic hirsutism groups were 16.04±1.4 and 7.32±6.85 µIu/ml, respectively (P<0.001). HOMA-IR values in PCOS and idiopathic hirsutism groups were 3.7± 3.36 and 1.67±1.75, respectively (P< 0.001). Mean of serum DHEA-S levels in PCOS and idiopathic hirsutism groups were 2.48±1 and 1.84±0.46 pg/ml, respectively (P< 0.001). These findings indicated a significant difference between the two groups. However, in terms of serum glucose, testosterone, androstendione, and follicular hormones levels, there were not any significant differences between the two groups. Conclusion: In both PCOS and idiopathic groups, there was a significant relationship between hirsutism and insulin resistance. This relationship, however, was more significant the PCOS group

Abstract

Background: Sodium arsenite has adverse effects on the reproductive system and vitamin E is a strong antioxidant and reproductive factor in the reproductive system. The aim of this study was to investigate the effect of vitamin E on the structure and the number of ovarian follicles during its development in rats treated with sodium arsenite. Materials and Methods: Pregnant Wister rats (n=4) were divided into 4 equal groups, including control, vitamin E (100mg/kg/day), sodium arsenite (8mg/kg/day), and sodium arsenite+vitamin E. Oral treatment of the pregnant rats started from the 7th day of pregnancy till the end of the weaning and continued till the age of 120 days. After the treatment period, the right ovary was removed and fixed, and then the mean number of ovarian follicles and atretic follicles, mean thickness of zona pellucida, and volume of oocytes and its nuclei in different types of follicle were determined. Data were analysed using one-way ANOVA and the mean difference was considered significant at P<0.05. Results: In the vitamin E+sodium arsenite group, significant increase(P<0.05) in the total number of follicles, mean number of primordial, primary, secondary, antral, and graafian follicles, significant reduction (P<0.05) in atretic follicles, and significant increase(P<0.05) in the thickness of zona pllucida in secondary and antral follicles as well as oocyte volume in parimordial and primary follicles and its nuclei in primary, secondary, antral, and graffian follicles were observed in comparison with the other groups. Vitamin E in the sodium arsenite+vitamin E group increased the number of different types of follicles and the thickness of the zona pllucida (P<0.05), decreased the number of atretic follicles, and increased the volume of oocyte and its nuclei to the level of the control group. Conclusion: Vitamin E can be administered as a supplement to compensate for the adverse effects of sodium arsenite.

Abstract

Background: Fennel (Foeniculum vulgare) has estrogenic, anti-inflammatory and antioxidant effects and traditionally used for treatment of infertile women. Polycystic Ovarian Syndrome (PCOS) is the most common endocrine disorder in women, which affects 5-10% of them in their reproductive age. This study has examined the effects of hydroalcoholic extract of Fennel on female Wistar rats with PCOS.

Materials and Methods: In this experimental study, 30 rats (200 ± 20 gr), was injected 2 ml of estradiol valerate subcutaneously and 6 rats were considered as control(intact). After 60 days, the rats were divided into control, sham and experimental groups. Rats in experimental group were treated with 250, 500, 1000(mg/kg BW) of intraperitoneal injections extract of Fennel for 10 days. Serum FSH, LH and Testosterone were measured by ELISA and serum levels of these hormones between treatments all groups were compared using One-Way ANOVA at a significance level of p<0.05.

Results: The data suggests that extract of Fennel can increase serum concentrations of FSH and decrease LH and Testosterone in treatment groups. As FSH is 500 and 1000 mg/kg levels of doses, LH and Testosterone in dose 1000 mg/kg have shown statistically significant differences compared to control groups.

Conclusion: Considering the increasing positive effect of extract of Fennel on serum concentration of FSH and decreaseing LH and Testosterone in rats with PCOS, this compound might be considered for treatment of the patients.

Abstract

Background: Polycystic ovary syndrome (PCOS) is a complex hormonal disorder affecting the reproductive and metabolic systems. The aim of this study was to investigate the effect of 8-week pilates training on anti-mullerian hormone level and cardiometabolic parameters in women with polycystic ovary syndrome.

Materials and Methods: In this semi-experimental study with pretest–posttest design, twenty PCOS patients (aged 28.16±4.29 y) were randomly assigned to pilates training or non-exercising control groups. The training group took part in an eight-week progressive pilates exercise program, three times weekly while the control group were instructed to maintain their normal daily activities throughout the eight-week experimental period. Serum levels of anti-mullerian hormone, body composition and metabolic parameters were assessed before and after the training period.

Results: After an 8 week pilates training, insulin resistance index, total cholesterol, LDL- cholesterol, triglycerides and abdominal fat were significantly decreased (p<0.05). Furthermore, anti-mullerian hormone concentrations decreased significantly in response to pilates training (p<0.05).

Conclusion: These results suggest that pilates training can have beneficial effects on metabolic indices and ovarian reserve in women with polycystic ovary syndrome over a two-month period.

Abstract

Background: Few studies have examined the effects of calcium plus vitamin D supplementation on metabolic profiles in overweight women with polycystic ovary syndrome (PCOS).This study was conducted to determine the effects of calcium plus vitamin D supplementation on metabolic profiles among overweight women with PCOS.

Materials and Methods: This randomized double-blind placebo-controlled clinical trial was conducted among 104 overweight and obess women with PCOS. Participants were randomly assigned into four groups to receive: 1) 1000 mg/d calcium+ vitamin D placebo; 2) 50000 IU/weeks vitamin D; 3) 1000 mg calcium/d and 4) calcium placebo + vitamin D placebo for 8 weeks. Fasting blood samples were taken at baseline and after 8 weeks intervention to measure metabolic profiles.

Results: After administrating calcium plus vitamin D supplementation, a significant decrease was observed in serum insulin (-3.3±11.4 vs.-1.1±8.5, -1.4±3.0 and +3.1±6.1 µIU/ml, p=0.03), insulin resistance (-0.7±2.4 vs.-0.3±2.5, -0.2±0.6 and +0.8±1.9, p=0.04), malondialdehyde (-0.6±0.3 vs.-0.1±0.2, -0.5±0.2 and +0.6±0.4 µmol/L, p=0.009) and a significant rise was seen in total antioxidant capacity (+35.2±21.7 vs.+22.5±37.7, +21.1±34.1 and -153.8±65.2 mmol/L, p=0.006) and glutathione (+216.0±79.3 vs.-47.5±60.2, +3.9±31.3 and -160.8±82.9 µmol/L, p=0.001) compared with vitamin D, calcium and placebo groups.

Conclusion: In conclusion, calcium plus vitamin D supplementation for eight weeks among women with PCOS had beneficial effects on metabolic profiles.

Abstract

Abstract

Background: We are aware of no study evaluating the effects of zinc supplementation on metabolic profiles of patients with polycystic ovary syndrome (PCOS). This study was conducted to evaluate the effects of zinc supplementation on glucose homeostasis parameters in PCOS women.

Materials and Methods: This randomized, double-blind, placebo-controlled trial was conducted among 52 women diagnosed with PCOS and aged 18-40 years old. Participants were randomly divided into two groups to receive 220 mg zinc sulfate (containing 50 mg zinc) supplements (n=26) or placebo (n=26) per day for 8 weeks. Fasting blood samples were taken at baseline and after 8 weeks of intervention to quantify glucose and insulin.

Results: After 8 weeks of intervention, zinc-supplemented patients had reduced fasting plasma glucose (FPG) (-4.3±9.6 vs. +0.5±6.0 mg/dL, p=0.03), serum insulin (-3.0 ± 2.9 vs. + 1.5 ± 8.4 µIU/ml, p=0.01), serum triglycerides levels (-15.6±40.3 vs. +14.5±25.3 mg/dL, p=0.002) and a significant increase in insulin sensitivity (+0.02±0.02 vs. -0.004±0.05, p=0.03) compared with the placebo.

Conclusion: Taken together, 220 mg zinc sulfate supplementation per day for 8 weeks among PCOS women had beneficial effects on metabolic profiles.

Abstract

Abstract

Background: Polycystic ovary syndrome(PCOS) is the most common endocrine aberration in women. PCOS is characterized by ovarian hyperandrogenism and anovulation resulted from a disorder of follicular maturation. Apoptosis is a regulatory mechanism for oocyte maturation and survival. Several studies have shown a possible role of Fas in ovarian apoptosis. The present study is the first investigation to examine the possible association of Fas rs1800682gene polymorphism with PCOS risk in Iranian women.

Materials and Methods: This case-control study was conducted on 251 patients with PCOS and 213 healthy control women. The Fas rs1800682 gene polymorphism genotypes were analyzed using the Tetra-ARMS-PCR method. Also, logistic regression analysis was used to investigate the association between genotypes and PCOS risk.

Results: There was a significant association between A allele and susceptibility to PCOS(OR =1.4, %95CI=1.08-1.83, p=0.011). Moreover, in the recessive genetic model for A allele, the AA genotype increased the risk of PCOS after adjusting age and body mass index(OR=1.6, %95CI=1.02-2.51, p=0.041).

Conclusion: For the first time, this study showed that Fas rs1800682 polymorphism is associated with PCOS risk in Iranian women and the A allele may act as a recessive allele for increasing the risk of PCOS.

Abstract

Background: We are aware of no study examining the effects of Soy supplementation on patients with polycystic ovary syndrome (PCOS).This study was conducted to evaluate the effects of Soy supplementation on metabolic profiles in women with PCOS.

Materials and Methods: This randomized, double-blind, placebo-controlled clinical trial was conducted among 70 women diagnosed with PCOS. Participants were randomly divided into two groups to receive 50 mg Soy isoflavone supplements (n=35) or placebo (n=35) for 12 weeks. Fasting blood samples were taken at baseline and after a 12-week intervention to quantify related factors.

Results: Intervention group, subjects who received Soy supplements had significantly decreased total serum testosterone levels (-0.2±0.4 vs. +0.1±0.3 ng/mL, p=0.01) compared with the placebo. In addition, a significant rise in plasma glutathione (+96.0±102.2 vs. +22.7±157.8 µmol/L, p=0.04) and a significant decrease in malondialdhyde levels (-0.7±0.8 vs. +0.8±2.3, p=0.001) was observed following the consumption of Soy supplements compared with the placebo.

Conclusion: According to this study, Soy supplementation among PCOS women had beneficial effects on metabolic profiles.

Abstract

Background and Aim: Polycystic Ovary Syndrome (PCOS) is one of the most common endocrine abnormalities that insulin resistance and obesity are among its main causes. The purpose of this study was to investigate the effect of chronic aerobic and resistive exercises along with supplementation of cinnamon on insulin resistance in women with polycystic ovary syndrome in Ahwaz in 2017.
Materials and Methods: In this semi-experimental study, 40 women with aged 18 to 30 years (BMI ≤24) were randomly divided into 4 groups. The intervention group performed 6 weeks, 3 sessions per week with intensity 50 to 75% of maximum heart rate for 50 to 85 minutes of exercise aerobic-resistance. Serum levels of insulin, glucose, and body composition indexes were evaluated. The t-test was correlated and ANCOVA was used.
Ethical Considerations: This study with research ethics code EE/ 96.24.3.77886/Scu.ac.ir has been approved by research ethics committee at Shahid Chamran University of Ahvaz, Iran.
Findings: Data analysis showed that body mass index, fat percentage and insulin resistance were decreased in exercise and supplementation exercises (p≤0.05). Cinnamon was also effective in combination with combined exercises in body composition and insulin resistance index (p≤0.05). Changes in fat free mass were not significant in any of the groups (p≤0.05).
Conclusion: It seems that this decrease in the difference in insulin resistance is more due to the combined exercise, and given the positive effect of cinnamon on reducing BMI, its use may be effective in the long term on insulin resistance.

Abstract

Background and Aim: Fennel with phytoestrogenic, antioxidant and anti-diabetic properties is used in traditional Iranian medicine to treat Polycystic Ovarian Syndrome (PCOS). Chronic inflammation is a new proposed mechanism in PCOS pathogenesis that suggests the role of immune system in this disease. Due to the main role of Treg cells in maintaining immune homeostasis, their impaired function, along with increased production of inflammatory compounds such as nitric oxide, can lead to persistent inflammation.
Methods & Materials: In this experimental study, 18 BALB/c mice were randomly divided into three groups including control group, untreated PCOS-induced group, and fennel-treated group. The control group received 0.1 ml sesame oil for 21 days. PCOS was induced by subcutaneous injection of Dehydroepiandrosterone (60 mg/kg/day) plus 0.1 mL sesame oil for 21 days. Mice in third group were treated by intraperitoneal injection of 500 mg/kg/day fennel extract after 21 days. The regulatory of T cells (Tregs) were analyzed by flow cytometry method and nitric oxide level determination was performed by Griess Reaction method. Ferric Reducing/Antioxidant Power (FRAP) assay was used to measure the total antioxidant power.
Ethical Considerations: This study was approved by the Ethical Committee of Arak University of Medical Sciences (Code: IR.ARAKMU.REC.1397.312).
Results: The estrous cycle was normalized after treatment with fennel. Fennel significantly reduced the number of ovarian cysts. Nitric oxide concentration decreased significantly while FRAP concentrations did not significantly alter. The number of Treg cells was also increased significantly.
Conclusion: Due to the role of immune system in the incidence of inflammation, concurrent modulation of both immune and endocrine systems may open new horizons in the design of new therapeutic interventions for PCOS.

Abstract

Background and Aim: In patients suffer from Polycystic Ovary Syndrome (PCOS), the secretion of the Luteinizing Hormone (LH) increases while adiponectin secretion and dopamine release decreases. Dopamine and adiponectin exert inhibitory effecs on LH secretion. In the present study the effects of L-dopa and dopamine receptor antagonists were investigated on LH secretion and adiponectin gene expression of in PCOS model rats to determine whether dopaminergic pathway might be involved in the decreasing LH via affecting adiponectin.
Methods & Materials: Following estradiol valerate- induced PCOS, fifteen PCOS rats were divided into 3 groups including saline receiving group, L-dopa(100 mg/kg) or simultaneous injections of sulpride(10 mg/kg), SCH23390 hydrochloride (1 mg/kg) and L-dopa(100 mg/kg), Five intact rats received saline as negative control group. Blood samples were collected via tail vein. Ovary and hypothalamus were dissected and frozen. Serum concentration of LH and relative gene expression of adiponectin in ovary and hypothalamus were determined by radioimmunoassay and real time-PCR method. 
Ethical Considerations: This study was approved by the Research Committee of University of Mohaghegh Ardabili (Code: 95.125.1). 
Results: Induction of PCOS caused a significant increase in mean serum concentration of LH and a significant decrease in mean relative gene expression of ovarian and hypothalamic adiponectin compared to control group. L-dopa caused a significant decrease in serum concentration of LH, a significant decrease in hypothalamic gene expression of adiponectin compared to PCOS rats. But it did not significantly increase ovarian adiponectin gene expression in comparison to PCOS rats. Dopamine receptor antagonists inhibit the effects of L-dopa on LH and hypothalamic gene expression of adiponectin.
Conclusion: Dopaminergic signaling pathway may be involved in decreasing LH secretion via increasing hypothalamic adiponectin gene expression level in PCOS rats.
