Abstract

Background: Giardiasis is the most common infectious diseases of medical parasitology and public health in our country and in many countries is important. Accourding the importance of treating the disease, particularly parasite resistance to drugs. Thus, the aim of present study was investigated the effect of gold nanoparticles on Giardia lamblia cyst stage of the in vitro.

Materials and Methods: In this experimental study, Giardia cysts from contaminated fresh feces were collected and concentrated with use 0.85 M sucrose methods for isolation of cysts. Samples after counting with Noubar slide were stored at 4°C. The gold nanoparticles at concentrations of 0.05, 0.1, 0.3 mg/ml prepared and verified, the particle size was determined. The effect of different concentrations of the nanoparticles at 5, 15, 30, 60 and 180 minutes was evaluated and compared to control groups (metronidazole). Data from the study were recorded and analyzed with T-test and Chi-square by used SPSS software.

Results: The results of the cytotoxic effect of nanoparticles on the different concentration and time of Giardia lamblia cysts in vitro compared with the positive control (Metronidazole) shows mean percent of effect gold nanoparticles increased with increasing concentration and exposure time, so that the concentration of 0.3 mg/ml from 62% at 5 min to 96% at 180 min increased (p<0.05). Also, the survey results show Giardia cysts with increasing exposure time , killing effect of gold nanoparticles at a concentration of 78% in 0.05 mg/ml to 96% at a concentration of 0.3 mg/ml greatest increased (p<0.05). As a result, the concentration of gold nanoparticles in comparison with Metronidazole has the most effect gold nanoparticles at a concentration of 0.3 mg/mL, almost the same effect of Metronidazole as the drug of choice in the treatment of Giardia lamblia.

Conclusion: Therefore, the results of this study, gold nanoparticles at a concentration of 0.3 mg/ml as an effective combination for killing Giardia lamblia cysts in vitro can be used. Thus, future studies on laboratory animals (In vivo) are recommended.

Abstract

Background: Maturation of oocytes and fertilization in vitro can be considered as one of the most important steps to treat infertility. In this study maturation medium was enriched with Platelet extraction (PL) which has high concentration of growth factors. Meiosis resumption and maturation was monitored after 18 hours of maturation.

Materials and Methods: In this experimental study, oocytes at germinal vesicle stage were obtained from mature NMRI female mice. Maturation medium was &alphaMEM and the control groups had 5-10% FBS and the medium in the experimental groups was enriched by 5, 10% PL and the combination of 5% PL and 5% FBS. Meiosis resumption and maturation were observed after 18 hours.

Results: The rate of matured oocytes in the experimental group 5% PL for both COC and DO group was significantly higher than the control groups (P<0.05). The maturation rate for 5% PL was 61% for the COC group and 72% for the DO group while this rate for 5% FBS control group was 53% and 50% respectively.

Conclusion: PL had a significant effect on meiosis resumption and maturation of oocyte at germinal vesicle stage. Based on these results, PL could be used as a maturation promoting factor.

Abstract

Background: Shigella is the causative agent of human shigellosis and its lipopolysaccharide is detected by TLR4. TLR4 belongs to Toll-like receptors family and many immunological pathways are triggered when these receptors are stimulated. Many researches showed increasing in TLR4 expression in mesenchymal stem cells through lipopolysaccharide treatment. The main goal of this study is detecting the optimum lipopolysaccharide between shigella strains through stimulation of immune system for vaccine studies.

Materials and Methods: In this experimental study human bone marrow derived mesenchymal stem cells were treated with three distinct concentrations (0.1, 0.01, and 0.001) of shigella (S. flexneri, S. dysenteriae, S. sonnei) extract containing lipopolysaccharide. Then TLR4 expression in mRNA level was investigated by RT-PCR and Q-PCR. The cells treated with phosphate buffered saline have been considered as a control group.

Results: Expression of TLR4 was shown in all of case groups except treatment with concentration 0.001 of extracts from sonnei and dysenteriae and also control group. The variations in the expression of TLR4 was dose-dependent in all of case groups. The maximum expression of TLR4 related to treatment with extract from shigella flexneri strain and the minimum expression related to treatment with shigella sonnei extract. The use of lipopolysaccharide from E. coli as a positive control indicated that lipopolysaccharide in shigella extracts is responsible for the increased expression of TLR4.

Conclusion: The TLR4 expression level was increasesed by S. flexneri extract, so it could be recommended for increasing vaccine efficiency.

Abstract

Introduction: Glyphosate is the most popular broad-spectrum herbicide globally due to the growing demand for glyphosate-resistant crops. Glyphosate exhibits harmful properties, including cytotoxicity, neurotoxicity, and reproductive toxicity. This study aimed to investigate the detrimental effects of glyphosate on ovarian histopathology in mice and the in vitro maturation (IVM) of oocytes following superovulation.
Methods: In this study, thirty-two female NMRI mice were randomly divided into the following groups: control, glyphosate, superovulation, and superovulation-glyphosate. Animals received glyphosate (0.5%) continuously through drinking water for three weeks. HMG and HCG were used to induce superovulation. Oocytes were collected from the ampulla, and the quantity and quality of oocytes were analyzed. Then, in vitro maturation (IVM) of oocytes was performed. At the end of the study, ovarian histopathology was analyzed.
Results: Compared to the control group, the glyphosate-treated group exhibited a significant decrease in secondary and Graafian follicles while demonstrating a concomitant increase in atretic follicles (P < 0.05). Additionally, the superovulation-glyphosate group showed fewer germinal vesicle breakdown (GVBD) and MII oocytes than the superovulation group. In the superovulation-glyphosate group, there was a notable reduction in GVBD and MII oocytes following in vitro maturation (IVM).
Conclusions: Glyphosate has the potential to damage ovarian tissue and adversely affect IVM and oogenesis.

Abstract

Introduction: The present study was conducted to investigate the effectiveness of acceptance and commitment-based therapy on the lifestyle of improving the health and emotional distress of women undergoing infertility treatment with the in vitro fertilization (IVF) method.
Methods: The method of this study was semi-experimental with a pre-test-post-test design with a control group and a follow-up stage. The statistical population of the study included women undergoing infertility treatment with the IVF method referring to the infertility center of Imam Khomeini Hospital in Tehran in the first six months of 2012. In this research, 27 women undergoing infertility treatment with IVF method were selected by voluntary sampling method and replaced by random method in test and control groups. The experimental group underwent acceptance and commitment therapy (10 sessions) for two and a half months. A health-promoting lifestyle questionnaire (HPLP) and an emotional distress questionnaire (Lavibond and Lavibond, 1995) were used as research tools. The data were analyzed using a mixed analysis of variance and Bonferroni post hoc test.
Results: It showed that the treatment based on acceptance and commitment to the lifestyle level improves health (Eta = 0.56, F = 19.74) and has a significant effect on the dimensions of emotional disorders such as depression (P < 0.0001, Eta = 0.58, F = 0.58). F = 34), anxiety (P > 0001; Eta = 0.59; F = 35.69), and stress (P > 0.0001; Eta = 0.61; F = 40.12) of women undergoing infertility treatment with the IVF, and has been able to lead to increase a healthy lifestyle, reduce depression, anxiety, and stress in these individuals.
Conclusions: Acceptance and commitment therapy, utilizing six psychological acceptance processes, cognitive breakdown, self under context, interaction with the present, specified values, and committed behavior, can be used as an effective treatment to reduce emotional distress in women undergoing IVF infertility treatment.
