Abstract

Background: Colorectal cancer is one of the most important and most common fatal types of cancer in the world. Identifying new agents which individually or in combination with other agents induce apoptosis in tumor cells is surely of great significance in treatment of colon cancer. The aim of the present study was to assess the effectiveness of applying lovastatin and α-tocopherol individually or in combination with each other in the induction of apoptosis in human colorectal cancer cells. Materials and Methods: In this trial, HT29 cells were exposed to various concenterations of lovastatin (5, 10, and 20 μmol) and/or alpha tocopherol (10, 20, and 25, and 30 μmol). After cell count, these cells were examined through trypan blue method and DNA fragmentation technique. Results: The findings of DNA fragmentation technique showed that each of the two drugs could induce apoptosis at all of the given concentrations. In the combination of 10 μmol concentration of lovastatin and 5 and 10 μmol concentrations of α-tocopherol, induction of apoptosis was not observed. Conclusion: Based on the extensive effects of statins, the concentration of lovastatin is seen as determining in its apoptosis function, and its combination with tocopheroles in high concentrations, by inducing apoptosis, can provide novel effective strategies for prevention of human colorectal cancer.

Abstract

Background: It can be used different models such as nonparametric, semi-parametric and parametric to estimation of survival. Depending on the nature of the data, some methods may be more appropriate. Using of appropriate model result more accuracy estimation of survival and it's affected. so the aim of this study estimation of survival and determination of its related risk factor. Materials and Methods: In this study, 169 patients with colorectal cancer who had treated in Hamadan MRI Center from March 2005 to Jun 2013 were evaluated. Survival time of patients is considered from diagnosis time to death or end of study. The effect of Demographic and clinical risk factors on survival is assessed by parametric model. follow up of patient Data were analyzed using Spss16 and R statistical software.

Results: The median survival was 4.3±40 months. Univariate analysis showed that the effect of staging of tumor, tumor grade, histology type of tumor, metastasis and relapse on survival were significant but in multivariate model histology type of tumor and metastasis were significant.

Conclusion: using of appropriate statistical model for detection of affected risk factor on patient with colorectal cancer can be prevented progression of disease and increased of survival of them.

Abstract

Background: Colorectal cancer is the most common gastrointestinal cancer. Investigating the factors that predict survival time for these patients is important.The purpose of this study was comparison of parametric models by estimating the prediction error and also identifying the effective factors on predicted survival time of patients with colorectal cancer.

Materials and Methods: This cohort study was conducted with 600 patients who were suffered from colorectal cancer in Taleghani Hospital of Tehran between 2001 to 2005 and they were followed up for at least 5 years. For identifying the effective factors on survival time, of the patients we analyzed the data by some parametric models such as Weibull, Exponential and Log logistic and compared these models with the estimation of prediction error by apparent loss method.

Results: Among 600 patients there was 344 men (57.3%) and 256 wemon (47.7%). Of total, 151 patients were died that 62.3% of them were men. Univariate analysis showed that the effect of BMI, sex, staging of tumor, tumor site were significant but in multivariate model staging of tumor and BMI were significant. By the estimation of prediction error, the best model was Log logistic.

Conclusion: With respect to the importance of survival time prediction, we found that we can use the prediction error to compare the parametric models. In addition, because of effectiveness of tumor stages and BMI in the patients’ survival time, survival time could be increased by an on-time diagnosis and an appropriate controled diet.

Abstract

Background: A small percent of patients with colorectal cancer (CRC) respond to 5-Fluorouracil (5-FU), as a first line of chemotherapy. In this study in, in order to design a new chemotherapy protocol, the effect of 5-FU and acriflavine (ACF) cotreatment on mortality rate of CRC cell lines was investigated.

Materials and Methods: Cytotoxicity of 5-FU and ACF against CRC cell lines (LS174T, SW480 and HCT116) was detected using MTT assay. Cells were treated with different concentrations of 5-FU (0.5-64 µM) or ACF (0.07-5 µM) for 72 hours and then cell viability and drugs IC50 was calculated. To assess the effect of ACF on anticancer activity of 5-FU, cells were cotreated with different concentrations of 5-FU and IC30 concentration of ACF.

Results: ACF and 5-FU suppress the viability of CRC cell lines in dose-dependent manner. 5-FU and ACF have most cytotoxic effect on LS174T and the lowest cytotoxic effect on SW480 cells. Cotreatment of ACF with 5-FU could not significantly change the sensitivity of cells against 5-FU (p>0.05).

Conclusion: In this study, the fatal and cytotoxic effect of ACF on three CRC cell lines was shown. However, cotreatment of ACF with 5-FU could not improve the anticancer activity of 5-FU.

Abstract

Abstract

Background: 4cure models are a model to analyze survival data that these models exist for long term survivors. Cure models are a special type of survival model where it is assumed that there are a proportion of subjects who had never event, thus, survival curve will eventually reach a plateau. Therefore, standard survival models are not appropriate because they do not account for the possibility of cure.The aim of the present research is to apply non-mixture cure model to analyze survival of patients with colorectal cancer.

Materials and Methods: We studied 232 patients with colorectal cancer who were visited and treated at Taleghani Hospital Research Center for Gastroenterology and Liver Disease in Tehran. These patients were diagnosed from 1987 to 2012 and followed up until 2013. The Effect of age, gender, family history, body mass index and site of infection were studied. Kaplan-Meier and Non-Mixture cure Model were used for analzing data.

Results: The ten-year survival rate after diagnosis in the studied patients was 64 % .A total of 60 (25.8 %) deaths due to colorectal cancer were observed. The mean of age at the time of diagnosis was 51.6 years. Based on non-mixed cure model, the rangs of age was 45-65 years old and BMI were significant.

Conclusion: When the population is divided into two groups (susceptible and non- susceptible individuals), using Cox semi-parametric model is not appropriate. Therefore, we should use cure models.

Abstract

Abstract
Background: Nowadays, drug resistances are the main problems in no response of cancer patients to drug. Identification of molecular mechanisms and causative agents of drug resistance can be important to determine the treatment method in different stages of disease. In this study, for the first time, expression of multidrug resistance (MDR1) gene was studied in colorectal cancer by Carbon Nanotubes (CNTs)-based nanobiosensor method.
Materials and Methods: At first, a nanobiosensor was designed based on carbon nanotubes (CNTs). After optimizing reaction condition to identify target DNA sequence, colorectal cancer patient’s cDNA samples were evaluated by nanbiosensor.
Results: After immobilizing the probe on CNTs, Fluorescence immersion was quenched but by adding complementary DNA, fluorescence again was observed. In hybridization reaction of cDNA with nanobiosensor high significant levels of fluorescence emission was observed in colorectal cancer tumor samples compared normal tissue indicating high level expression of MDR1gene in tumor tissue.
Conclusion: Finally, the evaluation of the expression of MDR1 gene by nanobiosensor indicated that in colorectal cancer tissue MDR1, mRNA level was higher than normal tissue. In addition, the results of this study indicated that carbon nanotubes-based nanobiosensor beside PCR-based method can be used as a very powerful tool for expression studies in human cancers at mRNA levels.

Abstract

Background and Aim: The protein encoded by the SGO1 gene is a member of the shugoshin family of proteins and protects the centromere during mitosis. lncRNAs are non-coding RNA with 200 nucleotides lengths, i.e., involved in regulating gene expression. The current study aimed to evaluate the expression of SGO1 and SGO1-AS1 in different stages of disease progression; we also compared their expression pattern in tumor tissues with healthy tissues in colorectal cancer patients.
Methods & Materials: In total, 40 tissue samples of patients with colorectal cancer were reported according to the examination and criteria with the approval of a pathologist. Besides, 40 normal tissues were sampled from a completely healthy part of the intestine of the same patients. After RNA extraction and cDNA synthesis, the Real-time RT-PCR technique was used to evaluate the expression of the desired genes in the study groups. ROC curve analysis was also used to determine the ability of each selected gene to diagnose the disease. 
Ethical Considerations: This study was approved by the Ethics Committee of Shahrekord Azad University (Code: IR.IAU.SHKREC.1398.020).
Results The obtained data suggested that SGO1 significantly decreased in the colorectal cancer tumor samples (P<0.001) and SGO1-AS1 LncRNA significantly increased expression, compared to adjacent healthy tissues. Additionally, in the age group of below 60 years, compared to the age group of over 60 years, SGO1 expression increased and SGO1-AS1 expression decreased. Based on the AUC obtained from the ROC diagram, it was found that the SGO1 gene with AUC=0.8041 and SGO1-AS1 with AUC=0.6364 could significantly distinguish a healthy population from patients with colorectal cancer.
Conclusion: According to the collected results, SGO1 -AS1 and SGO1 were significantly reduced and increased in tumor tissue, respectively; however, only the SGO1 gene was introduced as a good marker for diagnosing colorectal cancer.

Abstract

Background and Aim PBK is a mitogen-activated protein kinase (MAPKK) among MEK1/2 and MEK7 and can phosphorylate P38, JNK, and ERK in many cellular functions. The E2F transcription factor family also belongs to a class of cellular regulators acting as oncogenes and tumor suppressors. This study aims to investigate the expression of PBK and E2F7 in the early stages of colorectal cancer (CRC) compared to advanced stages based on the experimental and TCGA (The Cancer Genome Atlas) database.
Methods & Materials A total of 32 tissue samples of patients with CRC with the approval of a pathobiologist were collected according to the examination and criteria reported from different stages. After RNA extraction and cDNA synthesis, the RT-qPCR technique was used to evaluate the expression of the desired genes in the study groups. A receiver operating characteristic (ROC) curve analysis was also used to determine the ability of each of the selected genes to differentiate the two populations: stage I+II and stage III+IV.
Ethical Considerations In all stages of this research, codes of ethics of research and publication were observed.
Results In this study, it was shown that the expression of PBK and E2F7 significantly increased in stage I+II samples compared to stage III+IV. These data were confirmed by laboratory results and information extracted from the TCGA database. Also, based on the area under curve obtained from the ROC curves, these two genes are significantly distinguishable between stage I+II and III+IV populations in CRC.
Conclusion According to the results of this study, PBK and E2F7 genes are good markers in the diagnosis of CRC.
