Abstract

Introduction: Iodine  deficiency  disorders (IDD)  is  a  worldwide  health  problem.  Many  parts  of  the  Islamic  republic  of  Iran  had  been  known  as  areas  of  endemic  goiter.  IDD  was  accepted  as  a  priority  health  problem  in  the  country  and  a  National  IDD  Council  was  formed  in  1989.
Material  and  Method: One  of  the  main  strategies  of  National  Council for  IDD  control  was  to  provide  at  least  150mg  of  iodine  per  day  to  the  entire  population  through  making  available  iodized  salt.  This  study  was  performed  to  evaluate  the  National  IDD  council  program  in  1996, in  rural  and  urban  areas  of  Markazi  province,  1300  school  children, aged  8  to  10  years, including  50%  boys, were selected  through  random  sampling  Grading  of  goiter  was  performed  according  to  WHO. Classification, Serume, T3, T4  and  TSH  were  measured  by  RIA  and  urinary  iodine  by  digestion  method.
Results: The  mean  of  serume  T3, T4  and  TSH  were: 10±2.0 μg/dl, 160±28 ng/dl  and  2.3±1.3 μIu/ml  respectively.  There  were  no  differences  between  males  and  females  and  school  children  of  rural  or  urban  areas.  Four person  had  serum  T4  more  than  12.5μg/dl.  The  mean  urinary  iodine  was  16mg/dl  in  entire  population.  75%  had  urinary  iodine  more  than  10mg/dl. Only  11%  had  urinary  iodine  less  than  5μg/dl. Total  prevalence  of  goiter  was  35%  (36%  in  girls  and  34%  in  boys).
ConclusionBased  on  the  available  data, seven  years  after  distribution  of  iodized  salt, urinary  iodine  of  school  children  of  Markazi  province  does not  show  iodine  deficiency.  However  goiter  is  still  endemic  in  Markazi  province.

Abstract

Introduction: Preclampsia is a pregnancy-specific condition characterized by hypertention, proteinuria and generalized edema. The condition usually occurs in the third trimester and word wide is one of the most common causes of maternal mortality and morbidity. No study concerning the immunological etiology and pathogenesis of preclampsia has been done in Iran. The aim of this study was to evaluate the role of some immunological factors involved in the pathogenesis of preclampsia.
Materials and methods: This research was a clinical trial study. We measured C3, C4, IgG, IgA and IgM levels in 20 women with preclampsia, 20 normal pregnant women and 20 non-pregnant women matched for age, BMI, parity and gestational age. Seven patients had developed severe and thirteen mild preclampsia. After written informed consent was obtained, blood samples of subject group.
Collected serum collected was analyzed for immunological parameters by the single radial immunodiffusion (SRID) method of Mancini.
Results: C4 level was significantly decreased in women with severe preclampsia than in women with mild preclampsia and normal pregnancies, whereas C3 level did not differ between groups. IgG concentration was significantly higher in non-pregnant women than in women with preclampsia and normal pregnancies. There was a significant decrease in IgA concentration in normal pregnant women compared with non-pregnant women. There was a not significant difference between the IgM concentrations of both groups.
Conclusion: Our data show several changes in immunological factors in preclampsia which may contribute to the pathogenesis of the disorder in Iranian women.

Abstract

Introduction: Hidden hepatitis B is a condition in which the surface antigen of hepatitis B in the patient's serum is negative but the DNA of the hepatitis B virus is detected in the serum or liver tissue. In this study, the prevalence of latent hepatitis B in chronic hepatitis C patients and their biochemical and histological changes were investigated.
method: In this descriptive study, target sampling was performed so that 27 chronic hepatitis C patients whose HBsAg was negative and during 2001 and 2002 to two hepatitis centers of Tehran and Research Center for Gastrointestinal and Liver Diseases of Shahid University of Medical Sciences Beheshti came in and underwent liver sampling. On the hepatic paraffinic block of these patients, polymerase chain reaction tests were performed for the presence of HBVDNA, as well as immunohistochemical tests for the presence and detection of HBsAg and central hepatitis B antigen.
Results: Of the 27 PCR samples examined, patients reported positive HBVDNA in 5 cases (19%). In all of these patients, IHC tests were reported to be negative for HBsAg and HBcAg. Histological changes of cirrhosis and irreversible cirrhosis symptoms were seen only in the HBVDNA group.
Conclusion: The prevalence of latent hepatitis B is relatively high in patients with hepatitis C. In these patients, latent hepatitis B can exacerbate liver damage and accelerate the progression of cirrhosis.

Abstract

Background: Chronic hepatitis B virus (HBV) infection is a multi-factorial disease that is accompanied with serious clinical complications. Host’s genetic background, especially immune–genetic factors, is critical in the pathogenesis of infection. Gamma interferon ((INF-γ) and its receptor have an important role in immune response to the virus and clinical course of the disease. The aim of this study is to investigate the association between single nucleotide polymorphism -611G/A located in promoter of gamma interferon receptor1 gene (INFGR1) and chronic HBV infection. Materials and Methods: In this Case Control study, genomic DNA from peripheral blood samples of 150 chronically HBV infected patients and 150 healthy controls was extracted by phenol-chloroform method. DNA analysis was performed by PCR-RFLP method and P<0.05 was considered significant. Results: After stages of genotyping and statistical analysis, a significant difference was observed between patient and control group, so that genotype GG was higher in the control group compared to the patient group. Conclusion: The host’s immune-genetic background can play an important role in the pathogenesis of infectious disease. Variations in INFGR1 were related to several diseases. The results showed that the presence of GG allele is accompanied by a decrease in susceptibility to chronic HBV infection.

Abstract

  Background: In spite of designing and applying an effective vaccine against Hepatitis B virus (HBV), chronic infection with this virus is still one of the most important health problems worldwide. Host genetic background including single nucleotide polymorphisms play a significant role in chronicity or clearance of the infection. The final product of programmed cell death 1 gene (PDCD1) is expressed frequently on T-cells and in chronic viral infections, prevent the virus-specific T-cell response against the virus. In this study, the association of a single nucleotide polymorphism (+7146A/G) in intron 4 of PD1 gene with chronic hepatitis B infection in Iranian population has been assessed.

  Materials and Methods: 212 chronic HBV patients and 208 healthy controls were analyzed in this case-control study. Genomic DNA of the studied individuals was extracted and after performing polymerase chain reaction (PCR), polymorphism of +7146 was determined via RFLP method.

  Results: Frequencies of GG, GA and AA genotypes on position 7146 of the intron 4 of PD1 gene were 77.4%, 20.7% and 1.9% in patient group and 80.8%, 15.4% and 3.8% in control group, respectively. After statistical analysis, No significant difference was observed between patient and control groups (p=0.198).

  Conclusion: Genotype frequencies in the studied population are in accordance with the results of previous studies. Results of the present study suggest that there is not any association between A/G single nucleotide polymorphism in intron 4 of PD1 gene and susceptibility to chronic hepatitis B in Iranian population.

Abstract

Background: Irisin is an exercise-induced myokine that is reduced with type 2 diabetes and improves insulin resistance via the browning of white adipose tissues. However, irisin response to two types of exercise in patients with type 2 diabetes is unknown.

Materials and Methods: In this study, 22 diabetic Wistar rats (Induced by high-fat diet and injections Stz) were randomly assigned to 3 groups: high intensity interval exercise (HIIT), low intensity continuous training (LICT) and control (C). Both HIIT and LICT groups trained on the treadmill 5 sessions per week for 8 weeks. Blood samples were taken 24 hours after the last training session and plasma irisin, insulin and glucose levels were measured. ANOVA and Tukey post hoc tests were used to analyze data and the level of significance has been considered at p≤0.05.

Results: Data analysis showed that plasma irisin levels in the HIIT group were significantly increased compared to the control group (p<0.05). However, there was no significant difference between other groups (p>0.05). Plasma glucose in both HIIT and LICT groups was significantly decreased compared to the control group (p<0.05). But plasma insulin levels and insulin resistance index were not different between groups (p>0.05).

Conclusion: The results of this study showed that exercise training can increase plasma irisin in rats with type 2 diabetes. However, these changes are partially dependent on the type of exercise training.

Abstract

Abstract

Background: Human exposure to air pollution is associated with an increased risk of diseases such as heart failure, asthma and cancer. It has been suggested that oxidative stress is involved in air pollution-induced disorders. Recently, γ-glutamyltransferase (GGT) is known as a marker of oxidative stress. This study was aimed to evaluate the effect of exposure to outdoor air pollution on enzyme activity of GGT and also usage of GGT serum level as a marker for studying of harmful effects of air pollution in the resident with high air pollution level.

Materials and Methods: In this analytical cross-sectional study, 110 healthy adult men, never-smoking, who worked in an area with high air pollution and 90 men who worked in an area with low air pollution, as control group, were enrolled. All subjects were in the age range of 25-45 years with minimum work history of three years. The GGT activity in the serum samples was determined using a spectrophotometric method.

Results: Our results showed that the serum levels of GGT in the subjects in the areas with high air pollution (33.92 ± 1.61 U/L) did not differ significantly with those of control region (33.62 ± 1.74 U/L).

Conclusion: Overall, this study did not support the hypothesis that GGT enzyme could be considered as an oxidative stress marker following exposure to outdoor air pollution. Further studies with a larger sample sizes and also trials in other areas are required to confirm these results.

Abstract

Background: Parvovirus 4 (PARV4) was first discovered in 2005, in a hepatitis B virus–infected injecting drug user (IDU). To date, the best evidence about PARV4 transmission is parenteral roots and comes from IDU individuals. It seems that the prevalence of the virus in the normal population is very low. In this study, we investigated the prevalence of PARV4 virus among patients with chronic HCV infection compared with healthy controls and related risk factors among these groups.

Materials and Methods: A total of 206 patients, including 103 patients with chronic HCV infection and 103 healthy controls, were studied by use of nested-PCR and also real-time PCR techniques.

Results: AST enzyme levels with a mean of 40.45+34.84 and 18.58+5.9 in patients and healthy group respectively and the amount of enzyme ALT among patients with a mean of 40.45+35.75 and 21.50+11.35 in patients and healthy group respectively, were reported. Finally, after screening all DNA samples from patients and controls, we discovered that none of these people are infected with the PARV4 virus.

Conclusion: This study is the first to investigate the occurrence of PARV4 among HCV patients in Iran. The results show that, the virus is not important in Iranian population, even in patients with blood born infections such as HCV and further studies in other areas and various groups are required.

Abstract

Abstract
Background: The advantageous effects of the regular moderate physical activity (Exercise) on reproduction is widely accepted, but until now, the alterations of the expression of hypothalamic arcuate nuclei gene affecting on reproduction axis in response to this type of physical activity are not clear. Therefore, the goal of the present investigation was to study the effect of one –month regular moderate physical activity on neurokinin B gene expression and reproductive axis hormonal changes.
Materials and Methods: In the experimental study, fourteen adult male Wistar rats were randomly divided into control and one- month regular moderate exercise groups. After one - month physical activity (20m/min), the arcuate nucleus was isolated from brain and stored in -80 refrigerators for neurokinin B gene expression assay by Real-time PCR method. In addition, serum samples were taken to assess the corticosterone, luteinizing hormone and testosterone levels by ELISA method. Data were analyzed by Independent t-test in SPSS.
Results: Neurokinin-B gene expression level was lower in the group which received physical activity than the control group(p<0.05). Also, corticosterone serum concentration was decreased in the physical activity group(p<0.05). By contrast, the physical activity induced luteinizing hormone and Testosterone serum levels evaluation in exersice group compared with control group (p<0.05).
Conclusion: Regular moderate physical activity may improve male reproductive performance by reducing the corticosterone hormone level and decreasing neurokinin B expression.

Abstract

Background and Aim Among the plants with multiple healing properties and a long and common history of use in Iranian traditional medicine, is dog-rose plant with the scientific name of “Rosa canina” that belongs to the Rosaceae family. This study aimed to review the effectiveness of Rosa canina in treatment of different body organs and tissues and emphasizes its therapeutic mechanism.
Methods & Materials This is a review study. About 176 papers published from 2010 to 2018 were extracted from PubMed, Science Direct, Scopus, Google Scholar, Ovid, MagIran, and SID databases using related keywords and entered into study. 
Ethical Considerations Ethical issues (including plagiarism, misconduct, data fabrication, falsification, double publication or submission, redundancy) have been completely observed by the authors. 
Results Different studies had indicated the importance of Rosa canina as a medicinal plant protecting liver, lowering glucose and lipid levels, and having antioxidant and anti-inflammatory properties. The highest flavonoid activity in the Rosaceae family is related to Rosa canina. The use of herbal extracts in the treatment of common diseases, such as diabetes, fatty liver, urinary tract and Alzheimer's, has been recommended.
Conclusion High qualitative and quantitative diversity of effective ingredients, ease of access, absence of adverse effects of consumption and endemic nature of Rosa canina plant make it necessary to use it for the control and treatment of common human diseases.

Abstract

Background and Aim: Tendon is the extracellular matrix of the muscle that mechanically and structurally adapts to the mechanical load. However, the cellular and molecular mechanisms of this adaptation are not known yet. The purpose of this study was to investigate the effect of 6 weeks of resistance training on expression of two Transforming Growth Factor Beta 1 (TGF-β1) and myostatin genes in the tendon of fast- and slow-twitch muscles including Extensor Digitorum Longus (EDL) and Soleus (SOL).
Methods & Materials: Twelve male Wistar rats with 8 weeks of age were randomly divided into two groups of exercise (n=6) and control (n=6). The exercise group performed resistance training (Carrying weights with 40-160% body weight on the ladder) for 6 weeks, 5 sessions per week. Forty-eight hours after the last training session, all rats were sacrificed and the tendons of SOL and EDL muscles were extracted. The mRNA expression level of TGF-β1 and myostatin genes was assayed using real time polymerase chain reaction. Independent t-test was used for statistical analysis. 
Ethical Considerations: All experiments on animals were according to the ethical guidelines of Research Ethics Committee of Islamic Azad University, Central Tehran Branch (ethical code: IR.IAU.PS.REC.1398.296)
Results: The expression of TGF-β1 gene in EDL (P≤0.001) and SOL (P≤0.01) muscle tendons significantly increased, while the expression of myostatin gene in EDL (P≤0.001) and SOL (P≤0.05) tendons were significantly reduced. 
Conclusion: Resistance training appears to up-regulate the basal levels of TGF-β1 gene and down-regulate the basal levels of myostatin gene in tendons of fast- and slow-twitch muscles, where these effects are significantly more pronounced in the tendon of fast-twitch muscle.

Abstract


Background and Aim: Tendon, like the skeletal muscle, exhibits mechanical and morphological adaptations resulted from exercise training; however, little is known about the basic cellular and molecular mechanisms that regulate these responses. The aim of the present study was therefore to investigate the effect of 6 weeks of aerobic training on the TGF-β1, myostatin and MMP9 mRNAs expression in the tendon of fast- and slow-twitch muscles. 
Methods & Materials For this purpose, 12 male Wistar rats at 8 weeks of age were randomly divided into two groups: experimental (n=6) and control (n=6). The exercise group performed aerobic training for 6 weeks, 5 sessions per week. Forty-eight hours after the last training session, all rats were sacrificed and the tendons of soleus and Extensor Digitorum Longus (EDL) muscles were extracted. Expression of TGF-β1, myostatin and MMP9 mRNAs were assayed using RealTime-PCR. Independent t-test was also used for statistical analysis.
Ethical Considerations: All stages of the study were conducted according to the ethical guidelines and authorization of Research Deputy of Islamic Azad University, Central Tehran Branch No. IR.IAU.PS.REC.1398.296. 
Results: The results showed that the expression of TGF-β1 mRNA in EDL and soleus tendons significantly increased (P≤0.001), whereas the expression of myostatin in EDL tendon was significantly reduced (P≤ 0.001). Increased mRNA expression of MMP9 in the tendon of EDL and soleus muscles was not statistically significant (P>0.05).
Conclusion: It seems that aerobic exercise can modulate the expression of genes involved in the regulation of tendon collagen in a muscle type-dependent manner.
