Abstract

Background: Radiotherapy is one of the cancer treatment methods. Prescribed dose for each fraction is considered based on radiosensitivity of tumoral and normal tissues. Viral agents are the effiectiv factors on tissue sensitivity. This research aimed to determine the effect of ionizing radiation of Cobal 60 on radiosensitivity of Hela cells infected with Measles virus. Methods and Materials: In this study, the radiosensitivity of Hela cells is investigated experimentally and qualitively. The cells have been cultivated in two groups (experimental and blank) and plating efficiency has been obtained. Then 100λ measles virus with serial dilution method was used to induce infection in different ratio for experimental group. After cell growth and passage, the two groups were irradiated with 2Gy gamma radiation of cobalt 60. Results: Results respectively indicated cell death increases up to 5-7%, 15-20% and 50-65%, after 2Gy irradiation by Co 60 for contaminating to Measles in low, moderate and high concentrations. Conclusion: Radiosensitivity of tumoral cells increases when they are infected by viral agent. The result in radiotherapy of cancers showed, in prescribing dose fraction non cancer disease should be considered.

Abstract

Background: Vulvovaginal candidiasis is a common problem in women. The purpose of this study was to compare two identification methods new PCR analysis (with DNA extracted from samples) and conventional culture technique in detection of Candida species in vulvovaginal samples.

Materials and Methods: In this experimental-analytical study, 150 women of fertility ages participated and two vaginal discharge samples were collected by speculum. One sample used for direct DNA extraction as well as PCR and the other was used for culture and phenotypic evaluations. Phenotypic evaluations waere performed by germ tube and chlamydospore formation and specially culture in chrome agar medium to detect color of the colonies. PCR was performed by DNA extracted from samples as templates and finally size of Candida specific amplicons was determined.

Results: From 150 samples, 87 in culture and 127 in new PCR technique were positive. In culture method, from total 87  Candida species , 73 C. albicans, 12 C. glabrata and 2 C. tropicalis were isolated whereas in new PCR technique, from  total 127 candida species 107  C. albicans, 18 C. glabrata and 2 C. tropicalis were identified. Concordance of the two methods were calculated as 68 percent.

Conclusion: The new molecular method (innovative PCR) has the potential to rapidly and accurately diagnose Candida vulvovaginitis in patients and can be used for diagnosis of  Candida species in clinical specimens.

Abstract

Background: Nanoparticles due to their small size can overcome blood-testis barrier and affect spermatogenesis process. The aim of this study is to investigate the influence of trioxide (MoO3) nanoparticles on histological changes of testis and spermatogenesis process in adult male Wistar rats.

Materials and Methods: In this experimental study 30 adult male Wistar rats were randomely divided into five groups (n=6), including control, 2 sham groups, and 2experimental groups. Control group had no treatment. Two experimental groups received doses 5 & 10 mg/kg/BW nano molybdenum trioxide (20nm) respectively, and two sham groups received the same doses of normal saline by intraperitoneal injection. After 28 days, rats testis was removed and fixed in Bouin’s fixative for histological examination. The 5&mum sections were stained with hematoxilin-eosin.

Results: In experimental group which received 5mg/kg/BW nanoparticle, there was some disorganization of spermatogenic cells in some seminiferous tubules. In experimental group which received 10mg/kg/BW nanoparticle, a significant decrease was also observed in the number of spermatogenic and sertoli cells in comparison with the control group.

Conclusion: According to the findings of this study exposure to the high doses of (MoO3) nanoparticles can disrupt male reproductive system in a dose- dependent manner. Hence, the application of (MoO3) NPs should be carried out cautiously.

Abstract

Abstract

Background: Protein tyrosine phosphatase 1B (PTP1B) is a key enzyme in dephosphorylation of the insulin receptor (IR) and it is a central factor to induce the insulin resistance. The purpose of this study was to evaluate the effect of 12-week aerobic training on protein tyrosine phosphatase 1B gene expression and insulin resistance in diabetic rats.

Materials and Methods: In this study, 16 Wistar rats were divided into aerobic training and control groups. After inducing diabetes intra protaneally, aerobic training group performed training protocol for 12 weeks and 5 session/week. The duration and speed of each session increased progressively as 18 to 26 m/min and 10 to 55 min, respectively. Then, blood and tissue (from gastrocnemius) sampling were carried out in diabetic rats. Insulin resistance markers and PTP1B gene expression were evaluated by commercial kits and Real-Time PCR method, respectively.

Results: Findings showed that PTP1B significantly was decreased in diabetic rats of aerobic training group (p=0.0001). Also, glucose and insulin resistance significantly was decreased in aerobic training groups (p=0.02 and p=0.006, respectively). However, insulin in control rats was significantly increased (p=0.015).

Conclusion: It seems that, current aerobic training protocol has capability to decrease PTP1B and insulin resistance in diabetic rats. Furthermore, the direct correlation between PTP1B and insulin illustrated that any changes in insulin resistance due to exercise training associated with diminution of negative regulation of insulin signaling pathway.

Abstract

Abstract
Background: A lot of studies indicate that cancer chemotherapy drugs such as doxorubicin results in memory impairment. On the other hand, crocin as the chemical constituent isolated from the Saffron is effective on memory and motor enhancement. So, in this work, we have studied the co-administration effect of crocin and doxorubicin on avoidance memory and motor activity in adult male rat.
Material and Methods: In this study, 50 male rats were divided into 5 groups: control, sham, doxorubicin, crocin and treated rats with coadministration of doxorubicin and crocin. In crocin group, crocin injected 30mg/kg for 21 days and each rat in the chemotherapy group was treated once a week for 3 weeks with doxorubicin (5mg/kg). Treatment group, received doxorubicin and crocin at the same time. Sham groups administrated with saline. All drugs were injected intraperitoneally. After these procure passive avoidance memory, balance and exploratory behaviors were assessed respectively by shuttle box, rotarod and open field instruments.
Results: Memory in rats which have consumed doxorubicin significantly was decreased compared to other groups (p<0.001). Crocin treatment improved memory impairment following doxorubicin injection (p<0.001). Motor activity in open field and rotarod tests in treatment group showed significantly increased compared to doxorubicin group (p<0.001).
Conclusion: Crocin consumption beside of anticancer drugs such as doxorubicin has protective effect on the bad effects of chemotherapy drugs on memory and movement.
