Abstract
The total of IgM and IgA in serum and synivial fluids of 45 patients with Rheumatiod Arthritis (RA) are measured by critical method of ELISA. %93 of the patients had RF in their serum. The results show that there is no difference between the rate of IgMRF and IgARF in serum and synovial fluid. It was found that percentage Rf the ratio in serum is higher than that of synovial fluid by the calculation of Re titre to total immunogloboline. The fifference is significant from statistical point of view. The main places of RF production probably is in the joins of patient who are contracted.

Abstract
Introduction: different isotypes of antibody can be produced by immune system after antigen contact. Detection and measurement of different classes of antibody against the antigen is very important in some cases. The aim of this study is designing of an ELISA method on the basis of inhibition of enzyme activity by using a non-competitive inhibitor. Therefore in this study rheumatoid factor is used as a model for the detection of different other classes of antibodies against the antigen.
Materials and Methods: In this cross sectional analytical study, we measured IgM and IgA rheumatoid factors in sera of 10 patients with rheumatoid arthritis and positive latex test , by mixed and routine ELISA. In mixed ELISA the activity of the first conjugated enzyme was blocked by a non-competitive inhibitor after adding the substrate. Then the next conjugated antibody, which was specific for another isotype, was added. By optical density, results was comparisoned with routine ELISA.
Results: The obtained results showed that the average optical density is lower when compared with routine ELISA , but the difference is not statistically significant. however these two methods did not show any significant difference in quantifying antibody isotypes. Also there is a positive association between mixed and routine ELISA (r=0.9, p=0.001).
Discussion: Lower optical density in mixed ELISA is probably because of stick hindrance by the first conjugate. So, because there is no significant difference between the results of these two types of ELISA, and also no need to repeat the test for each isotype in this method, it is recommended to use the new method instead of the routine one to save time and reagents.

Abstract
Introduction: Peptic ulcer is a condition in which there is an ulcer in the stomach or duodenum. The incidence of peptic ulcer is 5 to 10% and is more prevalent in men. Infection with helicobacter pylori, NSAIDS, smoking and alcoholism are of the most important risk factors. Another risk factor is the O blood group. Since most research in this field are done abroad this study is done with the aim of determining the relation between ABO blood group and Rh with peptic ulcer. Materials and Methods: In this case-control study, 325 persons with dyspepsia were investigated. These individuals were referred for endoscopy. The case group consisted of 110 persons in whom endoscopy had revealed a peptic ulcer and control group 215 persons in whom endoscopy had showed no sign of ulcer. Blood grouping was performed by slide and tube methods .Chi square test was used for data analysis. Results: Based on the results, patients’ age was between 18-75 years. 68.6% of samples were women because they had more referrals. There was a significant relation between age and sex with peptic ulcer. 25.5% of patients were single and 72.5% married. Among all samples 32.9% had A blood group, 18.5% B, 11.1% AB and 37.5% O and 88% were Rh positive and 12% Rh negative. There was no significant relation between ABO blood group and Rh with peptic ulcer. The gathered data was analyzed using odds ratio, relative risk and Chi square test. Conclusion: Although some investigations have showed correlations between ABO blood group and peptic ulcer, our data showed no correlation. This may be due to the prevalence of helicobacter pylori infection among patients with peptic ulcer which needs more investigations. Other effective factors may be life style and nutrition.

Abstract
Background: Constipation is a functional gastrointestinal disorder and the use of probiotics is considered as a new solution in prevention and treatment of constipation. The aim of this study is to review the clinical evidence about probiotics effects in the prevention and treatment of constipation.
Materials and Methods: To review the accessible evidence about efficacy of probiotics in the prevention and treatment of constipation, the search of key words such as constipation, probiotic and yoghurt are conducted in databases Pubmed, Medlinplus and Cochran from 1980-2013 and of 49 articles about constipation, 16 yandomized controlled clinical trial articles and 3 pre-test and post-test randomized trials that investigated the effects of probiotics on constipation were included in this study.
Results: Probiotics produced lactic acid and fatty acids with short chains that decrease the colon pH and stimulate muscular contractions and intestinal peristalsis. Some of probiotics can increase the mucous discharge and change linked bile salts to free bile salts entering much water to colon and leads to soft defecation.
Conclusion: Considering studies, daily use of probiotics such as lactobacillus acidophilus and bifidobacteria lactic 105-1011 CFU/gr for at least 1-2 weeks can help to decrease constipation.

Abstract
Introduction: Salmonella infection (salmonellosis) is a common bacterial disease that affects the intestinal tract. Several methods like Multiplex or real-time PCR, ELISA, and Agglutination are used to identify these bacteria. However, normally rapid, cost effective and easy diagnostic methods such as agglutination test is recommended. In Iran, positive control antiserums used in diagnostic kits work based on polyvalent agglutination and are against O and H antigens. The purpose of this research was to produce specific anti-sera against O and H antigens for using in agglutination and ELISA kits.
Methods: New Zealand white rabbits were immunized by intravenous injections of inactivated bacterial O and H antigens adjusted to a cell density equivalent to a turbidity of a McFarland number 3 standard. Serum collection was performed 7 days after the last injection. Collected Antisera were tested with positive human specimens as well as cross-reaction antibodies. Absorption method was used to obtain specific anti-sera against O and H antigens. Produced Anti-O and Anti-H antibodies were mixed with bacterial H and O antigens respectively and incubated for 1 hours in 37˚c. The Mixture was centrifuged and the supernatant was collected. Furthermore, in order to use these antisera in specific kits such as ELISA, Immunofluorescence etc., purification methods like Ammonium sulphate precipitation, tangential Flow Filtration and Chromatography were performed. This study was approved by the ethics committee of Pasteur Institute of Iran (Code: IR.PII.REC.1399.006).
Results: Results of agglutination test before and after adsorption showed cross-reaction before adsorption and no cross-reaction with H and O antigens with monospecific antisera against O and H after adsorption, respectively. Moreover, high quality and quantity of mono-specific antibody was obtained after purification.
Conclusions: Serum-based assays are recommended for the timely diagnosis of the disease since these assays are specific, sensitive, inexpensive, and rapid. Therefore, the produced antiserum in the present research can be used in primary screening of salmonella infections based on agglutination tests which are cost effective and simple. In addition, purified anti-sera can be used in the development of ELISA and Immunofluorescence assays.

Abstract
Introduction: Several factors influence students' behaviors and develop motivation. Meanwhile, cognitive factors such as psychological capital play a more prominent role than others, and it is crucial to understand the factors that affect them. The current research purpose was to compare the effectiveness of mindfulness intervention and cognitive-behavioral game therapy on the psychological capital of female first-secondary school students in Amol City.
Methods: The research method was semi-experimental with a pre-test-post-test design with a control group. The statistical population included all the female students of the first secondary school of the seventh grade of the public schools of Amol City in the academic year of 2023. Among them, 45 people were selected by the available sampling method and were replaced randomly and by lottery in two experimental and control groups (30 individuals in the experimental group and 15 people in the control group). The people attending the first experimental group received mindfulness intervention (8 sessions) and two sessions per week for 60 minutes, and the second experimental group received cognitive-behavioral game therapy intervention (10 sessions) and two sessions per week for 30 minutes. The questionnaire used in this research included psychological capital. The study data was analyzed using the repeated measures analysis of variance (ANOVA) method.
Results: The findings showed that mindfulness and cognitive-behavioral game therapy affected the psychological capital of female students of the first secondary school in Amol City (P < 0.01). Also, mindfulness training on psychological capital was more effective than cognitive-behavioral game therapy (P < 0.01).
Conclusions: Although both methods of mindfulness training and play therapy improved students' psychological capital, the approach and method used in each are different. Mindfulness training focuses more on awareness and the presence of the mind in the present moment. At the same time, play therapy focuses more on strengthening psychological skills and abilities through interactive activities and games, which can better capitalize on increased psychological.

Abstract
Introduction: Being overweight is associated with increased cortisol secretion, along with heightened anxiety and depression. This study aimed to compare the effect of six weeks of concurrent moderate-intensity continuous training (MICT) and high-intensity interval training (HIIT) with HIIT training alone on body composition, plasma cortisol levels, and anxiety and depression levels in overweight male students.
Methods: In this semi-experimental research, 27 overweight students were selected and randomly divided into three groups 9: MICT+HIIT concurrent training, HIIT training, and a control group. The MICT )15 minutes of exercise per day at 60-65% of maximum heart rate with an additional 5 minutes every two weeks). The HIIT (3 minutes of running at 70-75% of maximum heart rate, followed by 3 minutes of active rest at 50% of maximum heart rate, with three repetitions in the first two weeks, four repetitions in the second two weeks, and five repetitions in the final two weeks).
Results: Both types of training protocols resulted in reductions in weight, body mass index, cholesterol, low-density lipoprotein, cortisol levels, anxiety, depression, and an increase in high-density lipoprotein (P < 0.05). There was no significant difference between the two types of exercise in terms of effectiveness on these variables; however, only peak oxygen consumption and triglycerides were associated with better changes in the MICT+HIIT group.
Conclusions: For overweight male adolescents aiming for weight loss and improved mental health, HIIT alone is sufficient, and adding MICT does not provide additional clinical benefits.
