Abstract

Introduction: Opioids abuse is a nationwide problem in our country. Regarding the potential of these drugs in crossing placenta they can cause many defects in fetuses of human and also animals. In the present study, the effects of maternal Morphine consumption on olfactory bulb development in rats have been investigated. Materials and Methods: Female Wistar rats (weight: 250-300g) were crossed with male rats and the zero time of pregnancy was detected. Existence of vaginal plug and also sperm in vaginal smear were indicators of mating and pregnancy. Experimental group received 0.05 mg/ml of Morphine in drinking water daily. On embryonic day 19, pregnant animals were anesthetized by Chloroform and embryos were taken out surgically. Weight of the embryos was determined by a digital balance and their length (Crown-Rump, Frontal-Occipital, Biparietal, Abdominal-Transfer and Anterior-Posterior diameters) were determined by a caliper. Then embryos were fixed in Formalin 10% and tissues were processed and sectioned and stained in Hematoxilin & Eosin and Bielschwosky staining methods. These sections were investigated for olfactory bulb development by MOTIC software and light Microscope. Data was analyzed using t-test. Results: The decrease in embryonic weight and their diameter for experiment group was significant. In microscopic study, the increase in thickness of mitral layer and the decrease in total number of Tufted, Mitral and Granular cells in experimental group were significant. Conclusion: This study showed that oral Morphine consumption leads to olfactory bulb defects.

Abstract

Background: Cutaneous leishmaniasis is a health problem in many areas of Iran. Cutaneous leishmaniasisis reported mostly in the central county of Qom province, including Ghanavat and Qomrood villages. This study was done to identify parasite species in human and rodents in order to illustrate the epidemiologic picture of the disease and provide an appropriate control program in 2010 Materials and Methods: This cross-sectional study was done on microscopic smears of 45 human samples and rodents samples hunted around Ghanavat and Qomrood villages in the central county of Qom province in 2010.In total,15 human samples and one hunted rodent sample were positive. In this study,the DNA of the parasites were extracted from the slides and analyzed by Leishmania specific premiers using ITS1 PCR-amplification (Internal Transcribed Spacer1). PCR (PolymeraseChain Reaction) products were digested by Haelll enzyme. Results: Overall, 15 human samples and one rodent sample from Merioneslibycus species were evaluated by PCR-RFLP (Restriction Fragment Length Polymorphism). After electrophoresis, it was demonstrated that the parasite was Leishmaniamajor in both human and rodent species. Conclusion: PCR-RFLP technique is an effective method to determine Leishmania parasite species in Geimsa stained slides from human and rodent reservoirs. One of the advantages of this technique is that it is possible to recognize the pathogen species of Leishmania parasite without gene sequencing. Besides, PCR-RFLP technique is a method of quite high sensitivity and specificity which can identify parasite species in addition to the diagnosis of leishmaniasis within 24 hours.

Abstract

Background: Retinoic acid (RA) is a vitamin A derivative and one of the most important inducing signals in vertebrates that is involved in differentiation, morphogenesis, apoptosis, and reproduction. This study was done to evaluate the role of RA in in vitro neural patterning of mouse embryonic stem cells (mESCs).

Materials and Methods: In this experimental study, upon formation, embryoid bodies (EBs) from mESCs, Royan B1, were induced by 1 µM RA for four days and then plated for eight days. Untreated EBs were considered as the control group. Finally, in both groups, neural induction and patterning of EB-derived neural cells were evaluated by using immunostaining, flowcytometry, and RT-PCR methods.

Results: RA induced neurogenesis in ES cells, from which 35% showed to express MAP2. RT-PCR analysis also indicated that RA-treated neural cells derived from ES cells could at the same time express Mash1, Pax6/7, and Dbx1/2 as dorso-ventral (DV) pattering markers and Hoxb4, Hoxc5, and Hoxc8 as the rostro-caudal (RC) axis markers.

Conclusion: RA induces in vitro neural induction along with neural patterning of ES-derived neural cells in DV and RC axes. Keywords: Mouse embryonic stem cells, neural patterning, retinoic acid

Abstract

Abstract
Background: According to application of Retinoic acid in differentiation of the stem cells to different cells and its role in apoptotic of cancer cells, the selection of appropriate dose for differentiation of stem cells is important. Thus in this study the effects of Retinoic acid in different concentrations on viability stem cells to select the appropriate dose for differentiation was investigated.
Materials and Methods: In this study, bone marrow mesenchymal stem cells were affected by different concentrations of Retinoic acid. Survival of cells was investigated after 3, 10 and 15 days of culture by MTT assay. DAPI staining was used to evaluate the number of apoplectic nuclei in treated cells after 10 and 15 days.
Results: After three days of culture, the results showed that a large number of cells are destroyed at concentrations of 10-4, 10-3 and 10-2M of Retinoic acid, while in 10-5 and 10-6 M of Retinoic acid, it is not observed many apoptosis. Amount of 10-5M Retinoic acid after 10 days showed significant apoptosis, while the concentration of 10-6 M Retinoic acid after 15 days showed significant apoptosis compared to the control group (p<0.05).
Conclusion: It looks that 10-6 M Retinoic acid is an appropriate concentration for differentiation of mesenchymal stem cells.

Abstract

Background and Aim: Ankle injuries are among the most common musculoskeletal injuries in sports, accounting for 10% to 15% of all injuries. Since controlling pain and inflammation in ankle sprains is one of the main goals of treatment, we decided to compare the therapeutic effects of Cheleh daghi Herbal ointment with a common therapeutic gel such as piroxicam.
Methods & Materials: This study was performed as a double-blinded clinical trial for 6 months from January 2019 to August 2019. After referral, all patients completed the consent form, and the patient's details and study variables were recorded by the evaluator in the research form. To measure joint inflammation, we measured the ankle with a meter tape in the ankle area (cm) and recorded. A goniometer was also used to measure the degree of ankle movements. The symptoms of ankle ecchymosis were also checked by physical examination and observation and recorded in the checklist. Statistical analysis was performed in SPSS V. 18 using statistical tests (the Mann-Whitney U test, the Chi-square, and the independent t-test).
Ethical Considerations: The Research Ethics Committee of Arak University of Medical Sciences approved the study (Code: IR.ARAKMU.REC.1396.55) and registered by the Clinical Trial (Code: IRCT2017071720258N53).
Results: The Mean±SD age in the control group was 34.2±8.6 years, and in the case group was 35.4±8.9 years. There was no statistically significant difference according to the Mann-Whitney U test (P<0.05). Also, the Mean±SD height in the control group was 1.7±0.12 m and in the case group was 1.72±0.11 m. The results also showed a statistically significant difference between the two groups studied before and after the intervention regarding the mean indicators of pain, swelling, and inflammation around the joints (P<0.05).
Conclusion: The results of our study showed that Cheleh daghi Herbal ointment for 3 weeks could be more effective in reducing local pain and inflammation than piroxicam gel treatment.

Abstract

Background and Aim: Older adults, particularly those living in nursing homes, are a vulnerable group whose poor nutritional status and inadequate nutrient intake provide the basis for the incidence of diseases and increase their health costs. The aim of the present study was to compare the Index for Nutritional Quality (INQ) between institutionalized (nursing-home) and non-institutionalized (community-living) older adults and to investigate its association with serum high sensitive C-reactive protein (hs-CRP) level in each group. 
Methods & Materials: In this case-control, age- and gender-matched study in Tabriz (East Azarbaijan, Iran), o subjects aged 65 years and older were recruited from nursing homes (n=76) and community (n=88). Anthropometric parameters were measured and body mass index (BMI) was calculated. A three-day record of the foods provided for the nursing home residents were weighed, using a digital scale. The whole food intake of older adults was converted to grams and dietary data were analyzed using Nutritionist IV software. Blood sample was drawn to measure serum hs-CRP level. SPSS software was used for data analysis.
Ethical Considerations: The present research was approved at Research Ethical Committee of Tabriz University of Medical Sciences (Code: TBZMED.REC.1394.152).
Results: In the present study, INQ for all nutrients (except for vitamins A, D, E, biotin, vitamin B6, folic acid, and iron) was significantly different between the two groups (P‌<0.05). The INQ<1 for these nutrients (except for iron) in both groups indicates insufficient intake of these micronutrients. However, INQ for nearly all nutrients (other than selenium) was higher in the community-living older adults, compared to nursing-home residents (P<0.05). The intake of minerals including calcium, magnesium, manganese, selenium, zinc, chromium, sodium, and potassium was also insufficient in both groups. Nursing-home residents did not meet their estimated needs for vitamins B2, B12, and copper. A significant reverse association was observed for serum hs-CRP level and INQ for vitamin D (P=0.04) and calcium (P=0.038) in the community-living older adults after adjusting for BMI. Such a correlation was also reported between INQ for vitamin B12 and serum hs-CRP level in both groups (P=0.02). 
Conclusion: Based on the findings of this study, it is recommended to pay more attention to the nutritional status of older adults, especially those living in nursing-homes. A diet rich in micronutrients, especially vitamin D, calcium, and vitamin B12 may help prevent the development of inflammation in old population.

Abstract

Background and aim: It is suggested that hearing impaired children with hearing aids and cochlear implant, showed poor pragmatic abilities. Pragmatic is the most related language component to social interaction. Further, it has effect on personality development and self-esteem. The main aim of the present study is investigating the relationship between pragmatic abilities and self-esteem in hearing impaired children.  
Materials and methods: Sixty hearing impaired children with hearing aids or cochlear implant were included into this study according to inclusion and exclusion criteria. Then parents or other caregivers were asked to fulfill Children’s Communication Checklist (CCC) and self-esteem questionnaire. The participants were divided into three groups based on hearing impairment severity, including: severe, moderate and mild. The relationship between pragmatic ability and self-esteem was examined in three groups and without considering categorization, by Pearson correlation coefficient, significant level was 0.05.
Ethical considerations: This project is approved in Arak university of medical sciences with ethical registration code IR.ARAKMU.REC.1399.150. All parents gave written informed consent form.
Findings: the results illustrated that there is significant relationship between pragmatic abilities and self-esteem in all hearing impairment children without considering categorization (p<0.05). By considering categorization, significant relationship was showed in self-esteem and pragmatic abilities in moderate and mild groups (p<0.05).
Conclusion: according to our analysis, pragmatic abilities and self-esteem are related. This relationship is more obvious in low severity impairment.
