Abstract

Background: Hypervariability of hepatitis C virus (HCV) proteins is an important obstacle to design an efficient vaccine for the infection. To construct a protective vaccine against HCV, a DNA vaccine containing conserved epitopes of the virus was designed. To enhance the induced immune responses, adjuvant activity of N-terminal domain of gp96 (NT(gp96)) was used.

Materials and Methods: A multi-epitope (PT) DNA vaccine encoding four HCV immunodominant cytotoxic T lymphocyte epitopes (HLA-A2 and H2-Dd) from Core, E2, NS3 and NS5B antigens in addition to a T-helper CD4+ epitope from NS3 protein and a B-cell epitope from E2 protein was designed and constructed. Then, NT(gp96) was fused to the PT DNA (PT-NT(gp96)). The stimulated cellular and humoral immune responses of PT and PT-NT(gp96) were evaluated in mice model.

Results: According to multicolor flow cytometry assay, the frequency of CD8+ T-cells producing IFNγ and TNFα in the splenocytes of immunized mice with PT-NT(gp96) (6.8%, 4%) was significantly higher than those of immunized with PT (0.9% , 0.8%), respectively. The same results have obtained in hepatic lymphocytes of the vaccinated mice. The level of IgG, IgG1 and IgG2a in the mice vaccinated with PT-NT (gp96) was significantly higher than the value obtained from the mice immunized with PT.

Conclusion: The results showed that PT DNA vaccine induces immune responses in mice model. Fusion of NT (gp96) to PT DNA vaccine causes to enhance cellular and humoral immune responses against HCV compared to sole PT vaccine.

Abstract

Abstract

Background: Acinetobacters are aerobic gram-negative bacteria which are distributed widespread in soil and water. The bacteria are isolated from cultured skin, mucous membranes, secretions and hospital environment. Acinetobacter baumannii, is a strain that more frequently isolated. Acinetobacter strains are often resistant against antimicrobial agents.

Materials and Methods: The method of this study was based on field, observation and test. On August and October 2015, samples were isolated from the soil and water of the Sadeghieh Square river in Tehran, respectively, and were transferred to the laboratory in the ice pack. 50 baumannii samples were isolated by biochemical methods (TSI, SIM, OF and gram test). November 1394, 100 clinical samples were isolated from Imam Khomeini hospital by biochemical method, and in the culture media Mueller Hinton agar plates were transferred to the laboratory. Antibiogram test for 150 baumannii samples was performed. Biofilms formation of Acinetobacter baumannii environmental and clinical samples was investigated by Congo red agar and culture plate methods.

Results: In all samples (clinical and soil), most of antibiotic resistance was 92% for imipenem and the resistance of water samples to imipenem was 99.9%. Biofilm formation by Congo red agar in water, soil, and clinical samles was resprctively 44%, 40% and 1%. All isolates were negative biofilm culture plate.

Conclusion: Considering Acinetobacter baumannii resistance to antibiotics and the lack of biofilm formation of in clinical and environmental isolates, it was concluded that there wasn’t any relationship between antibiotic resistance and biofilm formation.

Abstract

Background and Aim: Since late 2019, with the emergence of a new type of coronavirus that causes a new respiratory disease called COVID-19, there have been many concerns about the spread of this disease and how to deal with it. Due to the ability of the virus to be transmitted rapidly, diagnosing the infected individuals in the early stages for isolating them is critical. This study  aims to evaluate the reliability of Computed Tomography (CT) scan in diagnosing COVID-19.
Methods & Materials: Participants were 212 patients admitted to hospital with confirmed diagnosis of COVID-19. Demographic information, medical history, symptoms, and the chest CT scan results were collected and analyzed. Finally, the power of CT scans in the diagnosis of this disease was compared with the Real-Time Polymerase Chain Reaction (RT-PCR) molecular test.
Ethical Considerations: This study received ethical approval from the ethics committee of AJA University of Medical Sciences (Code: IR.AJAUMS.REC.1399.091).
Results: The sensitivity of CT scan in the diagnosis of COVID-19 was relatively high, but its false-positive results were also high.
Conclusion: CT scan is a relatively sensitive method for diagnosing COVID-19, but caution should be made due to its high false-positive results which can lead to increased financial burden on the health system.

Abstract

Background and Aim: The new Coronavirus Disease 2019 (COVID-19) outbreak initiated from China in late December 2019, has posed a great global health threat. Since patients with high blood pressure, diabetes, and cardiovascular diseases are at high risk of COVID-19, this study aims to investigate the prevalence, severity, clinical manifestations, and mortality rate in COVID-19 patients with underlying diseases compared to those without underlying diseases.
Methods & Materials: This study was performed on 1408 patients with COVID-19 admitted to Golestan, Hajar, Khanevadeh and Besat hospitals in Tehran, Iran. Required data including disease prevalence, severity, clinical manifestations and mortality rate in patients with and without underlying diseases were collected and then analyzed using statistical tests in SPSS software, considering a significance level of P<0.05.
Ethical Considerations: This study was approved by the AJA University of Medical Sciences (Code: IR.AJAUMS.REC.1399.091). Ethical principles have been observed in accordance with the guidelines of the National Ethics Committee and the COPE regulations.
Results: Patients were 912 men (64.8%) and 496 women (35.2%) aged 18-98 years (Mean age= 57.8 years). Results reported that 27% of patients were intubated and 21.9% were died. Blood oxygen level in 53.7% of patients was less than 93 mg/dL; 36.1% had a history of hypertension, 24.1% had a history of diabetes, and 21.7% had a history of cardiovascular disease.
Conclusion: The severity, clinical manifestations, and mortality rate of COVID-19 patients with cardiovascular disease, diabetes and hypertension are significantly different compared to those without underlying diseases.

Abstract

Background and Aim: In every age of history, human beings have been afflicted with the fear of disease. Undoubtedly, cancer is among the most concerning diseases; cancer pain is among the major pains. The present study aimed to compare the effects of cognitive-behavioral group therapy and mindfulness-based cognitive therapy on multidimensional pain symptoms in patients with breast cancer.
Methods & Materials: The present study was applied concerning the purpose and applied a quasi-experimental with pretest-posttest, a two-month follow-up, and control group design. The statistical population was all women with breast cancer referring to Tehran Cancer Research Center in 2018 who were selected by convenience sampling method, consisting of 30 patients. They were randomly divided into two groups of intervention and the control group. The repeated-measures Analysis of Variance (ANOVA) was used to analyze the obtained data. The research instrument was a multidimensional pain questionnaire (West Hyun-Yale) to evaluate multiple pain indices.
Results: The current study results suggested that cognitive-behavioral therapy and mindfulness-based cognitive therapy were effective on pain experience (P<0.001); however, there was no significant difference between the intervention groups respecting pain experience.
Conclusion Based on the present research findings, cognitive-behavioral therapy and mindfulness-based cognitive therapy impacted the management and pain intensity of patients with breast cancer.

Abstract

Background and Aim: The main objective of the current study was to compare the effectiveness of schema therapy and dialectical behavior therapy (DBT) on health-promoting lifestyle (HPL) and quality of life (QoL) of HIV/AIDS patients. 
Methods & Materials: The current study was an experimental study based on a pre-test-post-test design with a control group, and based on the objectives, it can be considered an applied study. The statistical population of the study included all HIV patients monitored by behavioral disorders clinics of the Healthcare Organization Centers in Hamedan County in Iran in 2019-2020. Using simple random sampling method, 45 patients were randomly selected from Shohada Clinic in Hamedan as the sample of the study, followed by randomly assigning them to three 15-member groups, i.e., two intervention groups and a control group. In order to measure the research variables, the Health-Promoting Lifestyle Profile (HPLP) (1987) and the Short Form (36) Health Survey (SF-36, 2001) were utilized. The obtained data were analyzed using ANCOVA in SPSS v. 12.
Ethical Considerations: The Ethics Committee of Hamedan University of Medical Sciences approved the study (Code: IR.UMSHA.REC.1398.1063). This study has been registered in the Iranian Registry of Clinical Trials (Code: IRCT20120215009014N348).
Results: The results of ANCOVA indicate that schema therapy and DBT affect HPL and quality of life, and there was a significant difference between the two groups (P‌<‌0.001). 
Conclusion: The results show that while both therapy methods affect the selected variables, schema therapy is more effective than DBT on HPL and Quality of Life (QoL).

Abstract

Background and Aim: The World Health Organization has recently identified COVID-19 as a pandemic. The outbreak of the disease has caused many people around the world to become extremely frightened and they show phobic signs. The purpose of the present study was to develop and normalize of Germophobia Questionnaire (GPQ).
Methods & Materials: The participants were 113 individuals (74-female) of the Iranian population. They completed the GPQ and Y-BOCS.
Ethical Considerations: All ethical principles and privacy policies of the results informed the participants. This study was approved in the second session of the Psychological Committee of the Islamic Azad University of Tehran’s province in March 2020.
Results: The results illustrate that the GPQ has acceptable internal consistency with Cronbach’s alpha coefficient (a=0.67), reliability coefficients with split-half (0.72), and one-week test-retest (r=0.73); in addition, the GPQ demonstrate moderate convergent validity with contamination obsessions checklist of Y-BOCS (r=0.44) as a convergent coefficient; and finally, the exploratory factor analysis indicates three-factors of the GPQ. 
Conclusion: Based on these findings, the GPQ has acceptable and good psychometric properties; researchers can use the GPQ as a reliable and valid instrument for assessing the germophobia levels in epidemic and pandemic crises like COVID-19.
