Abstract
Background: Live attenuated Shigella vaccines have shown promise in inducing protective immune responses in human clinical trials. The aim of this study was to design and construct pDS132::∆icsA as a suicide plasmid for targeted deletion of a region of icsA gene in Shigella. Materials and Methods: In this experimental study, species and serotypes of Shigella isolated from diarrhea samples of children at Firozabadi and Milad Hospitals of Tehran were confirmed by using serological and PCR tests. Identification primers of icsA gene were designed and then cloned to the pGEM-5zf vector and sequenced. According to icsA restriction enzyme map, 1751 bp of icsA gene was deleted by HincП restriction enzyme and the ∆icsA was constructed successfully. The pGEM∆icsA vector was digested by use of SphI and SalI enzymes and was then cloned to a suicide vector (pSD132). Precision of the process was confirmed by phenotype and genotype tests. Results: The Shigella dysenteriae type 1 strain was verified by serological tests and PCR. Sequence of the icsA gene in the native strain was identical to the strains submitted in the gene-bank database. Since the pDS132::∆icsA contains 1484 bp derived from icsA gene, it can be used to disrupt icsA gene as a specific suicide vector. Conclusion: Application of suicide systems facilitated mutant construction in more specific and effective methods in comparison with the other primary techniques such as serial passage.

Abstract
Background: Acanthamoeba is an opportunistic protozoan pathogen that is known to infect the cornea to produce eye keratitis and the central nervous system to produce lethal granulomatous encephalitis. The overall aim of the present study was to determine the anti-amoebic potential of natural compound Peganum harmala against the trophozoites and cysts of Acanthamoeba in vitro.
Materials and Methods: In this experimental study, a clinical isolate of Acanthamoeba was cultured and genotyped. The ethanolic extract of Peganum harmala was prepared. The trophozoites and cysts were collected by washing in page's saline. Various concentrations (1.25, 2.5, 5, and 10 mg/ml) of the ethanolic extract and polyhexanide 0.02% drop as positive control were tested at three different times (24, 48 and 72 h) on trophozoites and cysts of Acanthamoeba in vitro. The viability of trophozoites or cysts was tested by eozin method, MTT, and flowcytometry analysis.
Results: The results revealed that alcoholic extract had remarkable inhibitory effect on the proliferation of Acanthamoeba cysts as compared to non-treated control, and the inhibition was time and dose dependent. In the presence of 10 mg/ml ethanolic extract in medium culture after 72 h, no viable trophozoites were determined and 21.10% cysts of Acanthamoeba were viable. Percentage of trophozoites and cysts viability after adding polyhexanide 0.02% drop in medium culture after 72 hours was 0% and 23.71%, respectively.
Conclusion: Ethanolic extracts of Peganum harmala could be considered a new natural compound against the Acanthamoeba trophozoites and cysts. Further works are required to evaluate the exact effect of this extract on Acanthamoeba agents in animal models.

Abstract
Background and Aim This study aimed to investigate the clinical and laboratory characteristics of hydatid cyst disease in patients admitted to Imam Reza, Imam Hossein, and Bisotoon hospitals in Kermanshah City, Iran, from 2014 to 2018.
Methods & Materials In this descriptive study, we studied and analyzed the medical records of 350 patients with hydatid cysts who were hospitalized and underwent surgical treatment in Kermanshah hospitals from 2014 to 2018. Also, the demographic characteristics of patients, including gender, age, occupation, place of residence, and affected organs, were extracted and recorded.
Ethical Considerations The Ethics Committee of Kermanshah University of Medical Sciences approved this study (Code: IR.KUMS.REC.1398.366).
Results In total, 350 patients with a Mean±SD age of 43.2±19.7 in women and 43.3±20.7 in men were admitted with a diagnosis of hydatid cyst. The highest and lowest prevalence of hydatid cysts were in the age groups of 41-50 (16.5%) years and 1-10 (4.8%) years, respectively. The prevalence was 181 (51.7%) in women and 169 (48.2%) in men. The organs mostly infected with hydatid cysts were the liver, lung, and brain, with 241 cases (68.8%), 55 (15.7%), and 24 (6.8%), respectively.
Conclusion This study shows that hydatid cyst disease is an important disease, especially in rural areas of Kermanshah Province, Iran.
