Investigation of the Association between Polymorphisms of Factor V (G1691A) and Factor II (G20210A) with Recurrent Miscarriage in Iranian Patients

Background: The pathogenesis of recurrent pregnancy loss includes complex interaction of several genetic and environmental factors. Changes in blood coagulation factors during pregnancy may play an important role in the occurrence of recurrent abortions (RA). Recently, inherited thrombophilia has been considered as a possible cause. Therefore, in this study we have investigated association of factor V (G1691A) and factor II (G20210A) polymorphisms in Iranian patients with recurrent abortions.

Materials and Methods: A total of 203 women participated in this study: 105 women with two or more consecutive unexplained miscarriage as cases and 98 women with at least two healthy children as control group. Total genomic DNA was isolated from Peripheral blood leukocytes. The presence or absence of mutation in the FV (G1691A) and FII (G20210A) polymorphisms were assessed by PCR-RFLP, using Mnl1 and HindIII digestion enzymes, respectively. Finally, the data were analyzed using Chi-Square test.

Results: The results showed no statistical significant differences in the prevalence of FV (G1691A) and FII (G20210A) polymorphisms between patients and control group.

Conclusion: considering the results of this study, these polymorphisms Seem to have no role in etiology of recurrent pregnancy loss in the studied population.

Background: Breast cancer is the most common cancer and one of the main causes of cancer-related death all over the world that has become a major public health concern. Human apurinic/apyrimidinic endonuclease (ApE1) is a multifunctional protein that has an important role in the base excision repair (BER) pathway. Single- nucleotide polymorphism T>G found in exon 5 led to substitution of Asp>Glu at codon 148 (Asp148Glu). In this case-control study, we aimed to evaluate the association of this polymorphism on the risk of breast cancer in Guilan population.

Materials and Methods: To study gene polymorphism APE1 (Case- Control), blood samples were collected from 75 patients diagnosed with breast cancer and 75 control subjects, and genotyped by allele-specific PCR (AS-PCR). To estimate the association between genotype and allele frequencies in cases and controls, Chi-Square analysis was used.

Results: Analysis revealed no significant differences were found in genotype and allele distributions of ApE1 Asp148Glu polymorphism between breast cancer patients and controls (p=0.1, p=0.6 respectively) in this population.

Conclusion: Data from this study indicated no significant association between the Asp148Glu polymorphism and breast cancer risk (p=0.1). Further study is needed to clarify the impact of Asp148Glu polymorphism on the breast cancer.

Background: Cancer is one of the most common causes of deaths in the world. Cancer incidence and prevalence is increasing in the world. There is no clear trend is available on incidence of these cancers in Iran, therefore, this study aimed to determine the incidence of 10 common cancers among Iranian women from 2003 to 2009.

Materials and Methods: A cross-sectional study was conducted by analyzing existing data. Data from a national registry of cancer cases were received. Data included the records from the cancer registry system in Iranian women during 2003 to 2009. The incidence rates were then standardized using direct method. We used Cochrane Armitage Test for linear trend by Winpepi software to determine the variation in the incidence of cancers.

Results: The results show that the proportion of all cancers is rising in women as compared to all population. Breast, skin, colorectal, gastric, esophageal, thyroid, leukemia, ovary, brain and uterus were the most common cancer among Iranian women. During the years of the survey, these cancers are significantly increased.

Conclusion: The results of this study showed that the incidence of cancer in women is increased. screening programs can be useful in reducing the cancers.

Protective Effects of Imedeen on Spermatogenic Disorders Caused by Oxidative Stress Induction in Cyclophosphamide-Treated Mice

Background: One of the side effects of chemotherapy drugs is oxidative stress that can damage the sperm and decrease fertility potential. Antioxidant agents in Imedeen like Lycophence GS and Biomarine complex play important role in preventing the direct and indirect effects of free radicals. So, in this study, the inhibitory effects of Imedeen on the damage caused by cyclophosphamide were investigated.

Materials and Methods: In this experimental study, 60 mature male mice were divided into six groups. The control group received physiological serum, the second group received CP with 12mg/kg/day dosage, the third group received Imedeen with 111µg/kg/day dosage, the fourth group received Imedeen with 222 µg/kg/day dosage, the fifth group received CP and Imedeen with one dosage and the last group received CP and Imedeen with double dosage. Sampling and studies on sperm quality were performed after 35 days.

Results: The results obtained from the caudal epididymal sperm analysis revealed that treated with CP caused significant decrease in sperm count, motility, and viability, while abnormal sperms increased as compared to control gruop. These changes were associated with significant increase in DNA damage and chromatin abnormality in the caudal epididymal spermatozoa as evidenced by Acridine Orange and Aniline Blue staining respectively. Notably administration of Imedeen caused a considerable recovery in above-mentioned parameters.

Conclusion: The results suggest that Imedeen as an antioxidant could diminish the side effects of cyclophosphamide in the reproductive system of male mice.

Background: Phacoemulsification is a modern cataract surgery that developed surgery from large incisions into smaller incision size and lead to faster wound heal and earlier visual rehabilitation. The purpose of this study was to compare the outcomes and complications of 3.2 and 2.65 mm main Incisions in phacoemulsification .

Materials and Methods: In this clinical study in Amir Kabir Hospital of Arak, 78 patients with senile cataract were divided into two groups. Half of them experienced 3.2 mm main incision (first group) and the others were under main incision with 2.65 mm (group 2). Immediately after operation if patients had a wound leak, the standard stromal hydration was used and a suture was placed. A day after operation, the patients were examined in the light of iris prolapse, if it existed, then a suture would be palced.

Results: The occurrence of anterior chamber wound leak was 7.7% and 2.6% in group 1 and group 2, respectively. Stromal hydration was needed 5.1% and 3.2% in group 1 and group 2 respectively. In fact, suture was needed 2.6% and 0.0% in group 1 and group 2, respectively. There was no iris prolapse in both groups.

Conclusion: There was no statistically significant difference between two groups from the point of view of wound leak and the need for suture and the rate of iris prolapse (p>0.05). Also, in low or intermediate nuclear density grade, the smaller and the more permanent incision with 2.65 mm is preferred.

Background: Oxidative stress and antioxidant status may be associated with glaucomatous damage. The purpose of this study was to compare the serum antioxidant status between primary angle closure glaucoma patients and healthy subjects.

Materials and Methods: In this case-control study 66 primary angle closure glaucoma patients and 80 controls were enrolled. Peripheral blood sample obtained from patients. Superoxide dismutase(SOD) was assayed by inhibition the rate of adrenochrome formation. Catalase (CAT) was evaluated by decrease of H2O2 absorbance. Glutathione peroxidase (GPx) and Glutathione reductase (GR) were determined following NADP oxidation or reduction. Glutathione S-transferase (GST) was measured by increase in the absorbance of CDNB and glutathione conjugation.

Results: Antioxidant defense enzymes were significantly decreased in glaucoma patients over those of control groups. CAT (p<0.006), SOD (p<0.020) and GPX (p<0.004). A relative insignificant decrease of GR and GST activities was observed in glaucoma patients compared with healthy subjects.

Conclusion: The present study supports the hypothesis that oxidative stress is an important factor in the pathogenesis of glaucoma. Although primary angle closure glaucoma has an anathomical basis but decrease in antioxidant defense enzymes activities may have a role in pathogenesis of this type of glaucoma.

Background: Nowadays, the decline in birth rate is one of the most important social problems in developing societies. Infertility is defined as a failure to conceive in a couple trying to reproduce after one year of regular intercourse without contraception. Leptin have been implicated in maintaining normal female reproductive functions, including lactation, folliculogenesis, ovarian steroidogenesis, the maintenance of mammary gland morphology, the development of dominant follicles and oocytes, the maturation of the uterus endometrium, and menstrual cycle regulation. Sinyle-nucleotide polymorphism T>C found in exon 3 leads to substitution of Arg>Trp at codon 1.5 (R105W). In this case-control study, we aimed to evaluate the association of this polymorphism and the risk of female infertility in the population of Guilan.

Materials and Methods: Blood Samples were collected from 86 patients diagnosed with female infertility and 60 control subjects, and genotyped by allele-specific PCR (AS-PCR). To estimate the association between genotype and allele frequencies in cases and controls, Chi-Square analysis was used.

Results: Analysis revealed no significant differences were found in genotype and allele distributions of LEP Arg105Trp between infertility cases and controls (p=0.21, p=0.2) in this population.

Conclusion: Our findings indicated no significant association between the Arg105Trp polymorphism and female infertility risk (p=0.21). While, more studies are needed to confirm the results.

Comparing the Amount of EMG Activity of the Selected Involved Muscles in Ankle Strategy in Female Athletes While Standing on One Leg on Shuttle Balance and Wobble Board

Background: It seems that using shuttle balance which has recently been produced in Iran would be beneficial in exercise prescription for preventing sports injuries and recovery. The purpose of this study is comparing the amount of the electromyography activity of involved muscles in ankle strategy while standing on one leg on shuttle balance versus wobble board.

Materials and Methods: This study is a functional and cause-compare study. 15 female students 20-22 years of age having the enterance standards were selected meaningfully. The amount of EMG activity of selected muscles (Tibialis Anterior, Gastrocnemius, Rectus Femoris and Hamstring) was measured while standing on one leg on two devices. The difference in means of muscles activity in both of devices was estimated using multivariate analysis of variance.

Results: The results showed a significant difference between the amount of EMG activity of involved muscles (p=0.001). Also, the results of the intragroup effects showed that the electromyography activity of Tibialis Anterior, Rectus Femoris and Hamstring while standing on shuttle balance was significantly more than the activity while standing on wobble board (p<0.05). While the amount of activity on shuttle balance was higher as compared to wobble board, but there was no significant difference in Gastrocnemius EMG activity between two devices (p>0.05).

Conclusion: It seems that standing on shuttle balance can make higher electromyography activity in the muscles that are involved on ankle and thigh joints, i.e. Tibialis Anterior, Rectus Femoris and Hamstring. So it is recommended that shuttle balance can be used in balance training program.

Cloning of Fusion Protein Gene of Newcastle Disease Virus into a Baculovirus Derived Bacmid Shuttle Vector, in Order to Express it in Insect Cell Line

Background: Newcastle disease virus (NDV) is one of the major pathogens in poultry and vaccination is intended to control the disease, as an effective solution, yet. Fusion protein (F) on surface of NDV, has a fundamental role in virus pathogenicity and can induce protective immunity, alone. With this background, here our aim was to construct a baculovirus derived recombinant bacmid shuttle vector (encoding F-protein) in order to express it in insect cell line.

Materials and Methods: In this experimental study, at first complete F gene from avirulent strain La Sota of NDV was amplified by RT-PCR to produce F cDNA. The amplicon was cloned into T/A cloning vector and afterwards into pFastBac Dual donor plasmid. After the verification of cloning process by two methods, PCR and enzymatic digestion analysis, the accuracy of F gene sequence was confirmed by sequencing. Finally, F-containing recombinant bacmid was subsequently generated in DH10Bac cell and the construct production was confirmed by a special PCR panel, using F specific primers and M13 universal primers.

Results: Analysis of confirmatory tests showed that the recombinant bacmid, expressing of F-protein gene in correct sequence and framework, has been constructed successfully.

Conclusion: The product of this F-containing recombinant bacmid, in addition to its independent application in the induction of protective immunity, can be used with the other individual recombinant baculoviruses, expressing HN and NP genes to produce NDV-VLPs in insect cell line.
